摘要
目的应用气相色谱-质谱(GC-MS)技术对不同分化程度胃癌细胞系的代谢产物进行对比研究。方法通过GC-MS检测并比较中分化胃癌细胞系SGC-7901、低分化胃癌细胞系BGC-823和胃正常上皮细胞系GES-1的代谢谱特点,并筛选存在差异的代谢产物。结果本研究建立了正交偏最小二乘法-判别分析模型,此模型能够将不同分化程度的胃癌细胞系与胃正常上皮细胞系完全区分开。同时,在不同分化程度的胃癌细胞系与胃正常上皮细胞系之间发现了9种差异性代谢产物(变量权重值>1,P<0.05,相似度>700),分别为天冬氨酸、谷氨酸、天冬酰胺、腺苷、泛酸、γ-氨基丁酸、牛磺酸、肌氨酸和油酸。结论上述代谢产物有望成为潜在的胃癌标记物,对胃癌患者的诊断、治疗及预后监测具有重要意义。
Objective To compare and investigate metabolic profiles of gastric cancer cell lines in different degrees of differentiation by the gas chromatography-mass spectrometer(GC-MS).Methods Metabolic profiles characteristics of moderately differentiated gastric cancer cell line(SGC-7901)and poorly differentiated gastric cancer cell line(BGC-823)and normal gastric epithelial cell line(GES-1)were detected and compared by using GC-MS,and different metabolites were screened.Results Orthogonal partial least squares-discrimination analysis(OPLS-DA)models were established in this study to completely distinguish gastric cancer cell lines in different degrees of differentiation from a normal gastric epithelial cell line.At the same time,9 kinds of different metabolites were found between gastric cancer cell lines and a normal gastric epithelial cell line[variable important in project(VIP)>1,P<0.05,similarity>700],and they were aspartic acid,glutamic acid,asparagines,adenosine,pantothenic acid,γ-aminobutyric acid,taurine,creatine and oleic acid respectively.Conclusion The above metabolites may be potential biomarkers of gastric cancer,and they may have great significance in diagnosis,treatment and prognosis monitoring of patients with gastric cancer.
