摘要
在酸性条件下,用胃蛋白酶酶解瘤背石磺肌肉并提取胶原蛋白,采用正交试验法确定提取胶原蛋白的最佳工艺,利用SDS-PAGE、红外扫描、氨基酸组成分析方法综合分析瘤背石磺肌肉胶原蛋白的基本结构特征。实验结果显示,酶解温度为35℃,酶添加量8 mg/mL,酶解时间24 h时,提取率最大为73.09%。所制备的胶原蛋白由3条肽链构成,纯度高,几乎不含杂带,甘氨酸含量最高,约占25%,其次是特征氨基酸含量,有特征吸收峰,具有完整的三螺旋结构,且稳定性高,是典型的Ⅰ型胶原蛋白。因此,开发利用瘤背石磺作为胶原蛋白新来源具有研究价值,且为深入研究瘤背石磺肌肉胶原蛋白的功能性质提供数据基础。
Pepsin was used to hydrolyze the muscle of sea slug Onchidium struma and extract collagen in an acidic environment.The parameters of the extraction were optimized by orthogonal design.The structural features of collagen from the muscle of O.struma were analyzed by SDS-PAGE,infrared scanning and amino acid composition analysis.The results showed that the maximum extraction rate was 73.09%when the temperature of the enzyme was 35℃and the addition of enzyme was 8 mg/mL,and the time of the enzymatic hydrolysis was 24 h.The collagen was composed of three peptides,and was almost free of miscellaneous bands,the glycine content is the highest,about 25%.It was followed by characteristic amino acid content,with a characteristic peak of absorption.There was a complete three-spiral structure and is high in stability,and is a typical type I collagen.Therefore,it is of great value to develop as a new source of collagen,and to provide a data base for the further study of the functional properties from the muscle collagen of O.struma.
