益母草总生物碱对大鼠薄型子宫内膜VEGF和Ang-2表达的影响 被引量:14

Effects of total alkaloids of leonurus on expressions of VEGF and Ang-2 in rat thin endometrium

摘要 目的探究益母草总生物碱对大鼠薄型子宫内膜血管内皮生长因子(VEGF)和血管生成素2(Ang-2)表达的影响。方法选取40只6周龄SPF级雌性大鼠,随机分为4组,空白对照组、模型组、低剂量益母草总生物碱组、高剂量益母草总生物碱组,10只每组。除空白对照组外,其余3组采用宫腔注射无水乙醇建立薄型子宫内膜大鼠模型。建模后,低剂量益母草总生物碱组和高剂量益母草总生物碱组按照25 mg/kg和100 mg/kg的剂量灌服益母草总生物碱,空白对照组和模型组灌服等剂量生理盐水,灌胃1次/d,总共灌胃处理4 d。测量各组大鼠子宫内膜厚度;使用HE染色观察大鼠子宫内膜;使用免疫组化观察大鼠子宫内膜VEGF、Ang-2表达情况;使用Western blot检测大鼠子宫内膜VEGF、Ang-2蛋白表达情况。结果相比空白对照组,模型组大鼠子宫内膜厚度显著减小,VEGF、Ang-2蛋白表达显著减少,差异有统计学意义(P<0.01);相比模型组,益母草总生物碱组大鼠子宫内膜厚度显著增加,VEGF、Ang-2蛋白表达显著增加,且高剂量益母草总生物碱组显著高于低剂量益母草总生物碱组,差异有统计学意义(P<0.01)。结论益母草总生物碱可以增加薄型子宫内膜大鼠子宫内膜VEGF和Ang-2蛋白表达量,实现促进子宫内膜的血管生成,增加子宫内膜的厚度,且在一定范围内呈浓度依赖性。 Objective To explore the effects of total alkaloids of leonurus on expressions of vascular endothelial growth factor(VEGF)and angiopoietin 2(Ang-2)in rat thin endometrium.Methods A total of 406-week-old female SPF rats were selected.Rat models of thin endometrium were established by intrauterine injection of absolute ethyl alcohol,and were randomly divided into four groups,including blank control group,model group,low total alkaloids of leonurus(TAL)group and high TAL group,with 15 rats in each group.Low TAL group and high TAL group were given TAL at the doses of 25 mg/kg and 100 mg/kg respectively,and blank control group and model group were given the same dose of normal saline,and they were treated for once/d,with a total of 4 d of gavage.The endometrial thickness of rats in each group was measured.The endometrium of rats was observed by HE staining.The expressions of VEGF and Ang-2 in rat endometrium were observed by immunohistochemistry.The expression levels of VEGF and Ang-2 in rat endometrium were detected by Western blot.Results Compared with blank control group,the endometrial thickness in model group was significantly decreased,and the protein expression levels of VEGF and Ang-2 were significantly decreased(P<0.01).Compared with model group,the endometrial thickness in total alkaloids of leonurus groups was significantly increased,and the protein expression levels of VEGF and Ang-2 were significantly increased,and the indexes in high TAL group were significantly higher than those in low TAL group(P<0.01).Conclusion Total alkaloids of leonurus can increase the protein expression levels of VEGF and Ang-2 in endometrium of rats with thin endometrium,promote the angiogenesis in endometrium and increase the thickness of endometrium,and it is concentration-dependent within a certain range.
