发热伴血小板减少综合征疫苗株免疫原性比较

Comparison of immunogenicity of severe fever thrombocytopenia syndrome vaccine virus strains

摘要 目的比较重症发热伴血小板减少综合征(severe fever with thrombocytopenia syndrome,SFTS)疫苗株AH12和JS2011-013毒株的免疫原性及其疫苗原液的免疫原性。方法用灭活前滴度相同的AH12和JS2011-013株病毒免疫BALB/c小鼠,设佐剂组(铝佐剂终浓度为1 mg/mL)及非佐剂组,比较相同免疫剂量下,14、28及42 d诱导产生的中和抗体效价;用含有相同Gn抗原量的AH12和JS2011-013株疫苗原液免疫小鼠,比较相同免疫剂量下,14、28及42 d诱导产生的中和抗体效价。结果AH12和JS2011-013毒株以灭活前相同滴度或相同Gn抗原含量的条件下分别免疫小鼠,AH12毒株产生的抗体效价较高,免疫原性较强;佐剂的添加对于毒株免疫原性有增强作用。结论AH12毒株的免疫原性强于JS2011-013毒株。 Objective To compare the immunogenicities of severe fever with thrombocytopenia syndrome(SFTS)vaccine virus strains AH12 and JS2011-013 and the corresponding vaccine bulks.Methods BALB/c mice were immunized with AH12 and JS2011-013 strains containing aluminium adjuvant at a final concentration of 1 mg/mL or containing no aluminium adjuvant,at the same titers before inactivation.The neutralizing antibody titers 14,28 and 42 d after immunization with AH12 and JS2011-013 strains at the same dosages were compared.Meanwhile,the mice were immunized with the vaccine bulks prepared with AH12 and JS2011-013 strains at the same Gn antigen contents,of which the neutralizing antibody titers 14,28 and 42 d after immunization at the same dosages were compared.Results At the same titers before inactivation or the same Gn antigen contents,AH12 strain induced higher antibody titers in mice,indicating a high immunogenicity.The addition of adjuvant enhanced the immunogenicity.Conclusion The immunogenicity of strain AH12 was higher than that of JS2011-013 strain.
