摘要
采用高效液相色谱法测定中国沙棘、阜杂1号和杂优54号沙棘叶中没食子酸的含量,其色谱条件为:色谱柱为Amethyst C18-11(4.6 mm×250 mm,5μm);流动相甲醇∶0.1%磷酸为15∶85;流速1.00 mL/min;检测波长为273 nm。结果表明,中国沙棘叶中没食子酸含量为2.3633 mg/g,阜杂1号沙棘叶中没食子酸含量为0.8772 mg/g,杂优54号沙棘叶中没食子酸含量为1.6039 mg/g;没食子酸的进样量为5~60μg/mL时,与峰面积呈良好线性关系。沙棘叶中含有大量的没食子酸,其在抗癌药的开发方面具有广阔前景。研究结果可为沙棘的进一步开发利用提供依据。
In this experiment,the contents of gallic acid in Chinese seabuckthorn,Fuza 1 and Zayou 54 seabuckthorn leaves were determined by high-performance liquid chromatography.The chromatographic conditions were:Amethyst C18-11 colum(4.6 mm×250 mm,5μm),mobile phase methanol∶0.1%phosphoric acid(15∶85),flow rate 1.00 mL/min,detection wavelength 273 nm,column temperature 30℃.The results showed that the content of gallic acid in Chinese seabuckthorn leaves was 2.3633 mg/g,the content of gallic acid in seabuckthorn leaves of Fuza 1 was 0.8772 mg/g,and the content of gallic acid in seabuckthorn leaves of Zayou 54 was 1.6039 mg/g.There was a good linear relationship between gallic acid and peak area when the amount of gallic acid was 5-60μg/mL.Seabuckthorn leaves contain a lot of gallic acid,which has a broad prospect in the development of anticancer drugs.Research findings will provide a basis for the further development and utilization of seabuckthorn.
