摘要
目的:研究白藜芦醇(RSV)对血清剥夺诱导大鼠神经元自噬的影响及分子机制。方法:将原代培养的大鼠皮层神经元分为对照组(control)、血清剥夺组(SD)、RSV处理组(RSV)和RSV联合组SIRT1抑制剂EX527处理组(RSV+EX 527),利用乳酸脱氢酶(LDH)检测试剂盒检测细胞活性,免疫荧光染色检测沉默信息调节因子2相关酶1(SIRT1)的表达变化、Western Blot检测自噬相关蛋白LC3和Beclin1的表达水平变化。结果:与正常对照组相比,血清剥夺引起细胞活性下降、SIRT1表达下降,LC3-Ⅱ/LC3-Ⅰ比例减少,Beclin1表达降低(P <0. 05);经过RSV处理后,细胞活性增加(P <0. 05),大鼠神经元SIRT1表达增加,LC3-Ⅱ/LC3-Ⅰ比例增加,Beclin1表达增加;使用EX 527处理后,RSV的神经保护作用受到抑制。结论:RSV通过激活SIRT1活性促进大鼠皮层神经元自噬。
Objective: To study the effect of resveratrol(RSV) on autophagy of rat neurons induced by serum deprivation and its molecular mechanism. Methods: The primary cultured rat cortical neurons were divided into the Control group(control),serum deprivation group(SD),RSV treatment group(RSV) and RSV SIRT1 inhibitors EX 527 treatment group(RSV + EX527). LDH method was used to detect cell activity. Immunofluorescence staining was used to detect the expression changes of silent mating type information regulation 2 homolog-1(SIRT1). Western Blot was used to detect the expression change of autophagy LC3 and Beclin1 protein. Results: Compared with the control group,serum starvation resulted in decreased cell activity,decreased SIRT1 expression,decreased LC3-Ⅱ/LC3-Ⅰ ratio,and decreased Beclin1 expression(P < 0. 05). After RSV treatment,cell activity increased(P < 0. 05),SIRT1 expression in rat neurons increased,LC3-Ⅱ/LC3-Ⅰ ratio increased,and Beclin1 expression increased. The neuroprotective effect of RSV was inhibited after treatment with EX 527. Conclusion: RSV promotes autophagy of cortical neurons by activating SIRT1.
