摘要
目的研究荧光定量PCR(FQ-PCR)法检测外周血肺特异性X蛋白(LUNX)mRNA表达与肺癌微转移的关系。方法选择本院2012年3月至2017年8月诊治的225例非小细胞肺癌(NSCLC)患者作为研究组,30例肺部良性病变患者作为对照组。采用荧光定量PCR法检测两组患者外周血LUNX mRNA表达,并探讨其与肺癌微转移的关系。结果研究组患者LUNX mRNA表达阳性率为73.33%,明显高于对照组的10.00%(P<0.05);疾病分期高的患者表达阳性率高,Ⅱ、Ⅲ、Ⅳ期患者阳性率明显高于Ⅰ期患者(P<0.05);淋巴结转移为阳性患者表达阳性率明显高于淋巴结阴性患者(P<0.05);LUNX mRNA表达阳性患者2年生存率明显低于阴性患者(P<0.05),前者中位生存时间为14.27个月,后者中位生存时间为23.84个月。结论FQ PCR检测外周血LUNX mRNA表达可作为肺癌微转移的重要指标,对早期诊断微转移以及评估患者预后具有重要意义。
Objective To study the relationship between the expression of lung specific X protein(LUNX)mRNA detected by method of QRT-PCR in peripheral blood and micrometastasis of lung cancer.Methods 45 patients with non-small cell lung cancer(NSCLC)diagnosed and treated in our hospital from March 2012 to August 2017 were selected as the study group,and 30 patients with benign pulmonary lesions were set as the control group.The expression of LUNX mRNA in peripheral blood of the two groups was detected by fluorescence quantitative PCR and its relationship with micrometastasis of lung cancer was discussed.Results The positive rate of LUNX mRNA expression in the study group was 7.3.33%,which was significantly higher than that of the control group(10.00%,P<0.05),the positive rate of the patients with high stage of the disease was significantly higher than that of the patients with stage I(P<0.05),and the positive rate of the patients with positive lymph node metastasis was significantly higher than that of the patients with negative lymph node(P<0.05).The 2-year survival rate of patients with positive LUNX expression was significantly lower than that of patients with negative expression(P<0.05).The median survival time of the former was 14.27 months,while that of the latter was 23.84 months.Conclusion FQ-PCR can be used to detect the expression of LUNX mRNA in peripheral blood.It is an important index of micrometastasis of lung cancer and is of great significance for early diagnosis of micrometastasis and evaluation of prognosis.
