摘要
【目的】研究人参皂苷Rg1对载脂蛋白E基因敲除(ApoE^-/-)小鼠血脂、血管性血友病因子(von willebrand factor,VWF)蛋白表达及基质金属蛋白酶-9(matrix metalloproteinase-9,MMP-9)蛋白含量的影响。【方法】8周龄ApoE^-/-小鼠,高脂饲料喂养,随机分为3组(n=10):模型组、人参皂苷Rg1低剂量和高剂量组,每天分别腹腔注射生理盐水、30 mg/kg和60 mg/kg人参皂苷Rg1。8周后,全自动生化分析仪测定血脂水平变化;同时分别用免疫组化法和ELISA法检测胸主动脉VWF及MMP-9的蛋白表达。【结果】与模型组相比,人参皂苷Rg1低、高剂量组能显著降低ApoE^-/-小鼠血清总胆固醇(cholesterol total,TC),甘油三脂(glycerin trilaurate,TG),低密度脂蛋白胆固醇(low density lipoprotein cholesterol,LDL-C)(P<0.05);人参皂苷Rg1低、高剂量组胸主动脉VWF的蛋白表达和外周血MMP-9蛋白水平都显著降低(P<0.05)。【结论】人参皂苷Rg1能够通过调节ApoE^-/-小鼠血脂水平,保护血管内皮细胞,并下调MMP-9蛋白水平发挥抗动脉粥样硬化(atherosclerosis,AS)作用。
【Objective】To investigate the effect of ginsenoside Rg1 on serum lipids,the expression of Von willebrand factor(VWF)and Matrix metalloproteinase-9(MMP-9)in Apolipoprotein E knockout(ApoE^-/-)mice.【Methods】8 week old ApoE^-/- mice were fed with high-fat diet and randomly divided into 3 groups(n=10):model group,ginsenoside Rg1 low-dose and high-dose groups,Intraperitoneal injection of normal saline,30 mg/kg and 60 mg/kg ginsenoside Rg1 daily.After 8 weeks,the changes of serum lipid levels were measured by automatic biochemical analyzer.The protein expressions of VWF and MMP-9 in thoracic aorta were detected by immunohistochemistry and ELISA,respectively.【Results】Compared with the model group,ginsenoside Rg1 low and high dose group could significantly reduce serum TC,TG,LDL-C in ApoE^-/- mice(P<0.05);ginsenoside Rg1 low and high dose group thoracic aorta VWF Both protein expression and peripheral blood MMP-9 protein levels were significantly lower(P<0.05).【Conclusion】Ginsenoside Rg1 can protect vascular endothelial cells by regulating blood lipid levels in ApoE^-/- mice,and down-regulate MMP-9 protein levels to exert anti-atherosclerotic effects.
