摘要
泛素(ubiquitin,UB)是一种小分子质量蛋白。从氨基酸序列上看,杆状病毒UB与真核生物UB的差异体现在2段序列上,即第15~31个和第53~57个氨基酸,其余的序列高度保守。这种高度保守性使得许多商业化的抗泛素抗体难以特异地将二者区分开来。将人工合成的家蚕核型多角体病毒(Bombyx mori nucleopolyhedrovirus,BmNPV)UB(v-UB)多肽17TEPAETVADLKQ28作为免疫原免疫BALB/c小鼠,成功制备了针对该多肽的特异性单克隆抗体,实现了对v-UB的特异性检测。利用该抗体,确定v-UB在细胞质中形成的斑点状结构为P62内涵体(inclusion body)。获得的单克隆抗体为今后v-UB的研究提供了实验基础。
Ubiquitin(UB) is a kind of proteins with small molecule weight. The amino acid sequence alignment of UB showed that differences between baculovirual UB and eukaryotic UB lie in amino acid sequence 15-31 and 53-57. Whereas, other amino acid sequences remain highly conservative, which makes it difficult for many commercial antibo-dies against UB to distinguish baculovirual UB and eukaryotic UB specifically. In this study, BALB/c mice were immunized with synthetic polypeptide BmNPV UB(v-UB, 17TEPAETVADLKQ28) to prepare monoclonal antibody against v-UB, and then specific detection of v-UB was successfully achieved. With this antibody, it can be ascertained that dot distribution structure formed by v-UB in the cytoplasm was P62 inclusion body. Therefore, this monoclonal antibody can serve as an experimental material for future study on v-UB.
