基于超高效液相色谱-线性离子阱-静电场轨道质谱研究偏痛汤1号方的入血活性成分 被引量:5

摘要 目的研究偏痛汤1号大鼠给药后血中的主要活性成分。方法收集口服灌胃偏痛汤1号的大鼠血浆,采用超高效液相色谱-线性离子阱-静电场轨道质谱(UHPLC-LTQ-Orbitrap MS)技术进行检测,利用ACQUITY UPLC BEH C 18(2.1 mm×100 mm,1.7μm)色谱柱,以0.1%甲酸水溶液(A)-乙腈(B)为流动相系统进行梯度洗脱,样品数据经时间比对、精确分子量和离子碎片解析等综合分析。结果在偏痛汤1号方中共鉴定到14个入血成分,均为复方所含有的原型成分。结论此方法简单、稳定,适于偏痛汤1号入血成分的研究,所鉴定的14种血中移行成分是偏痛汤1号的体内直接作用物质,本研究结果将有助于阐明该复方作用的物质基础及体内作用过程。 Objective To study the main active ingredients in blood of No.1 piantong decoction medicated rats.Methods The rat plasma was collected after the lavage with No.1 piantong decoction.UHPLC-LTQ-Orbitrap MS technique was used in determination.The chromatographic column was treated with ACQUITY UPLC BEH C 18(2.1 mm×100 mm,1.7μm).The gradient elution was done with 0.1%formic acid-water solution(A)-acetonitrile(B)as the mobile phase system.The sample data were comprehensively analyzed by time comparison,accurate molecular weight and ion fragment analysis.Results A total of 14 incoming blood components were identified in No.1 piantong decoction and all of them were the prototype components in the compound formula.Conclusion This method is simple and stable and it is suitable for the study on the incoming blood components of No.1 piantong decoction.The identified 14 transitional components in blood are the objects on which No.1 piantong decoction works directly inside body.The study results are conductive to the explanation of the material basis of the effect of this compound formula and the mechanism in the body.
