摘要 为更好地控制黄芪精粉的质量稳定性,建立HPlC-ElSD法测定黄芪精粉中黄芪甲苷的含量的方法;采用安捷伦1260型高效液相色谱仪和Waters C18柱(4.6 mm×250 mm,5μm),以乙腈-水(32︰68)为流动相梯度洗脱,流速:1 mL/min,柱温30℃,蒸发光散射检测器检测,理论塔板数按黄芪甲苷峰计算不低于4000。结果表明,黄芪进样量在0.52~5.30μg内范围内均与峰面积呈良好线性关系,相关系数分别为:1.000,平均回收率分别为99.8%(RSD=0.59%),该方法方便快捷,重现性好,结果可靠,可用于测定黄芪精粉中黄芪甲苷的含量。 To better control the quality stability of Astragalus membranaceus powder,a method for determining the content of Astragaloside IV in Astragalus membranaceus powder by HPlC-ElSD method was established.Agilent 1260 high performance liquid chromatography and Waters C18 column(4.6 mm×250 mm,5μm),eluted with acetonitrile-water(32︰68)as mobile phase,flow rate:1 mL/min,column temperature 30℃,detected by evaporative light scattering detector,theoretical plate number according to astragaloside IV The peak calculation is not less than 4000.The results showed that the injection volume of Astragalus membranaceus was linear with the peak area in the range of 0.52~5.30μg.The correlation coefficients were 1.0 and the average recovery was 98.6%(RSD=1.9%).The method is convenient and fast.It has good reproducibility and reliable results.It can be used to determine the content of astragaloside IV in Astragalus membranaceus.
