木犀草素对喉癌Hep2细胞凋亡的影响 被引量:3

摘要 目的:研究木犀草素对喉癌Hep2细胞凋亡的影响及相关的作用机制。方法:磺酰罗丹明B(SRB)染色法检测0.5、1.0、2.0、4.0、8.0、16.0、32.0、64.0和128.0μmol/L木犀草素作用24、48、72 h后Hep2细胞存活率;Hep2细胞分别用0、10、20和40μmol/L木犀草素处理,流式细胞术检测各组Hep2细胞凋亡情况,Hoechst染色检测Hep2细胞中凋亡小体,Western blot实验检测凋亡相关蛋白Bax、Bcl-2、Bid的表达。结果:木犀草素呈时间浓度依赖性地抑制Hep2细胞存活(P<0.001.。木犀草素可以促进Hep2细胞的凋亡,诱导Hep2细胞产生凋亡小体,且能明显上调Bax、Bid蛋白的表达,降低Bcl-2蛋白的表达(P均<0.001.。结论:木犀草素通过上调Bax、Bid蛋白的表达和降低Bcl-2蛋白的表达诱导Hep2细胞的凋亡。 Aim:To study the effects of luteolin on the apoptosis of laryngeal cancer Hep2 cells and the mechanism.Methods:The survival of Hep2 cells treated by different concentrations(0.5,1.0,2.0,4.0,8.0,16.0,32.0,64.0,128.0μmol/L)of luteolin after 24,48 and 72 hours was detected by SRB staining.The effects of different concentrations(0,10,20,40μmol/L)of luteolin on apoptosis of Hep2 cells was detected by flow cytometry.Hoechst staining was used to detect the apoptotic body in Hep2 cells.The expressions of Bax,Bcl-2,and Bid proteins were detected by Western blot.Results:Luteolin significantly inhibited Hep2 cells survival in a time- and concentration-dependent manner( P <0.001.. Luteolin induced apoptosis and apoptotic bodies were observed in Hep2 cells. Western blot showed that luteolin increased the expressions of Bax and Bid, while reduced the expression of Bcl-2( P <0.001.. Conclusion :Luteolin induces apoptosis of Hep2 cells by upregulating Bax and Bid proteins and downregulating Bcl-2 protein.
