下调肝星状细胞中SOX12的表达可抑制肝硬化 被引量:1

摘要 目的研究SOX12在肝星状细胞与正常肝细胞中的表达,并探讨SOX12通过介导TGF-β1(转化生长因子β1)的表达在调节肝星状细胞的活性中对肝硬化的影响。方法培养细胞LX-2(肝星状细胞)与Changliver cell(正常肝细胞),使用PCR及Western blot检测细胞中SOX12与免疫组化检查肝组织中肝硬化各项指标的表达,同时探讨SOX12与TGF-β1之间的关系;用细胞转染的方法研究抑制SOX12基因的表达后对TGF-β1的影响。结果1)SOX12蛋白在人类肝细胞和星状细胞中均有高表达;2)抑制SOX12基因的表达可以使肝星状细胞中TGF-β1以及collagenⅠ(胶原蛋白Ⅱ)表达量减少;3)SOX12可调控肝星状细胞的活性和TGF-β1的表达,导致细胞外基质的积累。结论下调肝星状细胞中SOX12的表达可抑制肝硬化。 Objective To investigate the expression of SOX12 in hepatic stellate cells and normal hepatocytes,and the effect of SOX12 on liver cirrhosis by mediating the expression of TGF-β1 in hepatic stellate cells.Methods LX-2(hepatic stellate cells)and Changliver cell(normal hepatocytes)were cultured.PCR technique was used to detect the expression of SOX12 and immunohistochemistry was used to detect the expression of various indexes in liver cirrhosis.At the same time,the relationship between SOX12 and TGF-β1(transforming growth factor)was explored out.Inhibition of SOX12 by cell transfection to gene expression on TGF-β1 was examined.Results 1)SOX12 protein was highly expressed in both human hepatocytes and stellate cells;2)the expression of TGF-β1 and collagenⅠin hepatic stellate cells were decreased through inhibiting expression of SOX12 gene;3)SOX12 regulateed the activity of hepatic stellate cells and the expression of TGF-β1,resulting in the accumulation of extra-cellular matrix.Conclusions Down-regulation of SOX12 expression in hepatic stellate cells inhibits liver cirrhosis.
