摘要
为了研究VHA-c3基因在拟南芥响应非生物胁迫中的作用。通过农杆菌介导,将设计的dsDNA片段整合在拟南芥基因组上,筛选获得了能够不同程度特异沉默VHA-c3基因的9个转基因株系c3-1~c3-9,并对VHA-c3基因沉默株系进行ABA和葡萄糖胁迫处理。在ABA和6%葡糖糖处理后,c3-1株系主根的相对伸长量和种子的萌发率都有一定程度的抑制,但是,其被抑制效果远远低于野生型株系。并且,随着VHA-c3基因沉默效率的增加,沉默株系受ABA或葡糖糖的抑制效果越不明显;非损伤微测技术测定结果表明,ABA促进了c3-1株系的H+内流能力。综上,VHA-c3基因沉默株系表现出对ABA抑制作用不敏感的现象。
In order to elucidate the roles of VHA-c3 gene against abiotic stress in Arabidopis thaliana,the designed double strand DNA fragments was integrated into the genome of Arabidopsis thaliana by Agrobacterium mediation,and then nine transgenic lines c3-1~c3-9 was screened to specifically silence the coding gene of VHA-c3 subunit.It was found that under the stress of ABA or glucose,the relative root length and the germination rate of seeds of c3-1 line did not decreased obviously as wild strain.Moreover,with the increase of VHA-c3 gene silencing efficiency,the silent lines were not significantly inhibited by ABA and glucose.Non-invasive micro-test technique was used to measure the ability of ABA to promote H+influx in c3-1 lines.In conclusion,VHA-c3 silenced lines showed insensitivity to the inhibition of ABA.
