祛风活血丸对实验性自身免疫性葡萄膜炎大鼠血清IL-15、IFN-γ表达的影响 被引量:3

Qufeng Huoxue Pills Has Effect on Expression of IL-15 and IFN-γ in serum of Rats with Experimental Autoimmune Uveitis

摘要 目的:观察祛风活血丸对实验性自身免疫性葡萄膜炎(Experimental autoimmune uveitis,EAU)大鼠血清白细胞介素-15 (Interleukin-15,IL-15)、干扰素-γ(Interferon-γ,IFN-γ)的影响。方法:将80只(160眼)雄性Lewis大鼠根据随机数字表法分为空白组、模型组、祛风活血丸常规剂量组、祛风活血丸2倍剂量组、熊胆开明片组,每组16只。除空白组外,其余大鼠进行主动免疫,建立EAU大鼠模型。模型制备成功后,连续灌胃给药14天,ELISA法检测大鼠血清中IL-15、IFN-γ的相对表达量。结果:与空白组比较,模型组各时间点IL-15、IFN-γ明显升高(P<0.01);与模型组比较,祛风活血丸常规剂量组、祛风活血丸2倍剂量组以及熊胆开明片组IL-15、IFN-γ的表达均降低(P<0.05,P<0.01)。结论:祛风活血丸可能通过抑制促炎症因子IL-15和IFN-γ的表达发挥其抗炎作用,从而达到改善EAU病理过程的效果。 Objective:To observe the effect of Qufeng Huoxue pills on interleukin-15(IL-15) and interferon-γ(IFN-γ) in serum of rats with experimental autoimmune uveitis(EAU).Methods:A total of 80 male Lewis rats(160 eyes) were divided into the blank group,the model group,the routine-dose Qufeng Huoxue pills group,the double-dose Qufeng Huoxue pills group and the Xiongdan Kaiming tablets group according to the random number method table,16 rats in each group.Except those in the blank group,other rats were given active immunization to establish EAU rat models.After successful modeling,the rats were administrated by gavage for 14 days;the relative expression of IL-15 and IFN-γ in serum was detected by ELISA method.Results:Compared with those in the blank group,IL-15 and IFN-γ at each time point in the model group was significantly increased(P<0.01);compared with that in the model group,the expression of IL-15 and IFN-γ in the routine-dose Qufeng Huoxue pills group,the double-dose Qufeng Huoxue pills group and the Xiongdan Kaiming tablets group was decreased(P<0.05,P<0.01).Conclusion:Qufeng Huoxue pills may exert its anti-inflammatory effect by inhibiting the expression of proinflammatory cytokines including IL-15 and IFN-γ,thereby improving the pathological process of EAU.
