摘要
目的探讨血清淀粉样蛋白A(Serum amyloid A,SAA)在咪喹莫特银屑病样小鼠中可能的作用。方法 24只小鼠外涂5%咪喹莫特乳膏或凡士林62.5 mg,1次/d,连续6 d,然后分为4组,每组6只。1组局部凡士林组腹腔注射控制性IgG,2组局部凡士林组腹腔注射抗-SAA抗体,3组局部咪喹莫特组腹腔注射IgG,4组局部咪喹莫特组腹腔注射抗-SAA抗体。所有小鼠在2周后安乐死,并取背部皮肤进行后续检测。结果咪喹莫特银屑病样小鼠皮损中SAA mRNA表达明显增加;中和SAA后咪喹莫特银屑病样小鼠皮损的厚度变薄,真皮浸润的炎症细胞减少,皮损中白细胞介素(IL)-17A和趋化因子20 mRNA的表达减少。结论咪喹莫特银屑病样小鼠皮损中SAA的表达明显增加,中和SAA可减轻银屑病相关的增生和炎症。
Objective In order to explore the possible role of serum amyloid A(SAA) in imiquimod psoriasis-like mice.Methods Twenty-four mice were treated with 5% imiquimod(IMQ) cream 62.5 mg or vaseline(Vas) once daily for six days,then they were divided into four groups with six cases in each group. Group one was treated with topical vaseline with intraperitoneal injection of control IgG;group two was treated topical Vaseline with intraperitoneal injection of anti-SAA antibody;group three was treated topical IMQ with intraperitoneal injection of control IgG;group four was treated topical IMQ with intraperitoneal injection of anti-SAA antibody. All mice were euthanized after two weeks and the back skins were taken for subsequent testing. Results SAA was significantly expressed in the lesions of imiquimod-like psoriasis mice. After neutralizing SAA,the thickness of imiquimod psoriasis-like mice was thinner,and the inflammatory cells of dermal infiltration were reduced. Interleukin-17 A and Chemokine 20 mRNA expression was also reduced. Conclusion SAA was highly expressed in skin lesions of imiquimod-treated psoriasis-like mice,and neutralizing SAA reduced psoriasis-associated hyperplasia and inflammation.
