摘要
目的探讨微小RNA(miRNA)-27、八聚体结合转录因子4(Oct4)、性别决定区Y框蛋白2(SOX2)在口腔鳞状细胞癌(OSCC)组织中的表达及其与患者临床特征和预后的关系。方法选取116例OSCC患者的OSCC组织和40例口腔良性病变患者的口腔良性病变组织。采用实时定量聚合酶链反应(PCR)检测miRNA-27的相对表达量,采用免疫组织化学染色法检测Oct4和SOX2蛋白的表达情况。比较OSCC组织和口腔良性病变组织中miRNA-27、Oct4蛋白、SOX2蛋白的表达情况,以及不同临床特征OSCC患者的miRNA-27、Oct4蛋白、SOX2蛋白表达情况。分析miRNA-27、Oct4蛋白、SOX2蛋白表达与OSCC患者预后的关系,采用Cox比例风险回归模型分析OSCC患者5年生存率的影响因素。结果OSCC组织中miRNA-27的相对表达量明显低于口腔良性病变组织(P﹤0.01),OSCC组织中Oct4蛋白、SOX2蛋白的阳性表达率均高于口腔良性病变组织(P﹤0.05)。不同吸烟史、分化程度、TNM分期、T分期、N分期的OSCC患者OSCC组织中miRNA-27的低表达率比较,差异均有统计学意义(P﹤0.05);不同吸烟史、TNM分期、N分期的OSCC患者OSCC组织中Oct4蛋白的阳性表达率比较,差异均有统计学意义(P﹤0.05);不同TNM分期OSCC患者OSCC组织中SOX2蛋白的阳性表达率比较,差异有统计学意义(P﹤0.05)。OSCC患者的5年生存率为62.93%(73/116)。miRNA-27低表达患者的5年生存率明显低于miRNA-27高表达患者,miRNA-27低表达+Oct4阳性表达+SOX2阳性表达者患者的5年生存率明显低于其他患者,差异均有统计学意义(P﹤0.01)。Cox比例风险回归模型分析结果显示,有吸烟史、TNM分期为Ⅲ期、N分期为N2~3期均是OSCC患者5年生存率的独立危险因素,而miRNA-27高表达是OSCC患者5年生存率的独立保护因素(P﹤0.05)。结论OSCC组织中miRNA-27的相对表达量较低,而Oct4和SOX2蛋白的阳性表达率均较高,miRNA-27、Oct4蛋白、SOX2蛋白有望成为预测OSCC患者预后的辅�
Objective To explore the expression of microRNA-27(miRNA-27),octamer binding transcription factor4(Oct4),sex determining region Y-box 2(SOX2)in oral squamous cell carcinoma(OSCC)tissues and their relationship with clinical characteristics and prognosis of OSCC patients.Method A total of 116 OSCC tissue specimens from patients with OSCC and 40 oral mucosa tissue specimens from patients with benign oral disease were collected.Real-time quantitative polymerase chain reaction(PCR)was utilized to detect the relative expression of miRNA-27,and immunohistochemical assay was employed to determine the expression of Oct4 and SOX2 protein.The expression of miRNA-27,Oct4 protein,and SOX2 protein in OSCC tissues and benign oral disease tissues were compared,besides,the expression of miRNA-27,Oct4 protein,and SOX2 protein in OSCC patients with varied clinical characteristics were also assessed.The association of miRNA-27,Oct4 protein,and SOX2 protein with the prognosis of OSCC patients were analyzed,furthermore,the factors affecting 5-year survival rate of OSCC patients were examined using Cox proportional hazard regression model.Result The relative expression of miRNA-27 in OSCC tissues was lower than that of benign oral disease tissues(P<0.01).In OSCC tissues,higher positive expression rates of Oct4 and SOX2 protein were observed as compared with benign oral disease tissues(P<0.05).There were significant differences regarding low expression rate of miRNA-27 in patients with OSCC in terms of smoking history,degree of differentiation,TNM stage,T stage,and N stage(P<0.05);the positive expression rate of Oct4 protein in OSCC patients with different smoking history,TNM stage,and N stage were significantly different(P<0.05);moreover,the positive expression rate of SOX2 protein in OSCC patients with different TNM stage was significantly different(P<0.05).The 5-year survival rate was 62.93%(73/116)among patients with OSCC.The 5-year survival rate of the patients with low expression of miRNA-27 was less than that of patients with hig
