摘要
以长白山东方红林场和汪清兰家林场采集的笃斯越桔叶片为样本,采用PCR方法扩增psbA-trnH基因,与GenBank中的越桔属psbA-trnH序列进行对比分析,分析psbA-trnH基因序列在越桔属植物间的序列差异。结果表明:越桔属psbA-trnH序列遗传距离的范围区间为0.000~0.013,平均遗传距离为0.005。草地越桔共有3个有效鉴别位点,分别在第36碱基位的碱基为C,在第289碱基位插入了1段TATTCATTTTG碱基序列,在第398碱基位的碱基为T,蔓越桔共有2个有效鉴别位点,分别在第195碱基位的碱基为C,第339碱基位缺失1段TTGAAAATAA碱基序列,狭叶越桔共存在3个有效鉴别位点,分别在第8碱基位点缺失1个碱基,第169位点碱基为T,第428位点碱基为A,乌饭树在第288碱基位为T,红豆越桔在第236位碱基缺失,鹿莓越桔第305位点碱基为T。草地越桔与蔓越桔与其他越桔属植物的碱基序列对比存在明显的差异。通过Neighbor-Joining系统发育树分析得知,笃斯越桔与红豆越桔亲缘关系较近。越桔属植物的psbA-trnH序列存在属间差异,故可以用来作为鉴定越桔属植物的DNA条形码。
The psbA-trnH gene was amplified by PCR using the leaves of the Vaccinium uliginosum collected from the Changbai Mountain Dongfanghong Forest Farm and the Wangqing Lanjia Family Forest Farm, and compared sequence differences with the psbA-trnH sequence of Vaccinium in GenBank. The results showed that the range of genetic distance of the psbA-trnH sequence of Vaccinium was 0.000-0.013, and the average genetic distance was 0.005. There were 3 effective identification sites in the Vaccinium pratense, the 36 th base was C, and a fragment of TATTCATTTTG was inserted at the 289 th position, the 398 th base was T. There were two effective identification sites in Vaccinium macrocarpon, the base at position 195 was C, and a fragment of TTGAAAATAA was deleted at the 339 th position. There were three effective identification sites in the Vaccinium angustifolium, a single base was deleted at the 8 th base position, the base at position 169 was T, and the base at 428 was A. The Vaccinium bracteatum was T at the 288 th position, the Vaccinium vitis-idaea was deleted at the 236 th base, and the base 305 of the deerberry bilberry was T. There was a significant difference in the base sequence comparison between Vaccinium pratense and Vaccinium macrocarpon and other Vaccinium plants. The Neighbor-Joining phylogenetic tree analysis showd that the Vaccinium uliginosum was closely related with the Vaccinium vitis-idaea. The psbA-trnH sequences of Vaccinium plants were different among genera, so it could be used as DNA barcode for identification of Vaccinium plants.
