摘要
目的:研究白藜芦醇改善丙硫氧嘧啶诱导大鼠甲状腺肿的作用。方法:筛选丙硫氧嘧啶造模剂量,将造模成功大鼠随机分为对照组、左甲状腺素钠8μg/kg组、白藜芦醇0. 2、0. 4 g/kg组,另设正常对照组。各组给予相应药物或溶媒,连续灌胃6周,取各组大鼠甲状腺,称重,生化仪检测血清中FT3、FT4、总T3和T4、TSH的含量;HE、Masson染色观察大鼠甲状腺病理学变化;观察白藜芦醇对大鼠甲状腺形态参数的影响。结果:5. 0 mg/kg的丙硫氧嘧啶能够诱导大鼠甲状腺显著肿大;与正常对照组比,模型组大鼠甲状腺重量显著增大,FT3、FT4、总T3和T4含量降低,TSH的含量升高,HE染色结果显示模型组大鼠甲状腺滤泡增生,滤泡腔较小,滤泡内胶质大量减少,Masson染色结果显示模型组胶原纤维大量增生。大鼠上皮细胞高度(HFE)明显增高,而泡腔体积(AFC)显著降低。与模型组比,不同剂量的白藜芦醇给药作用后甲状腺重量明显减轻,FT3、FT4、总T3和T4含量升高,TSH的含量降低,HE染色结果显示大鼠甲状腺滤泡减少,滤泡内胶质显著增多,Masson染色结果显示胶原纤维大量减少,大鼠上皮细胞高度升高,AFC降低。结论:白藜芦醇对丙硫氧嘧啶诱导的大鼠甲状腺肿具有一定的改善作用。
Objective: To investigate the effect of resveratrol (Res) on the improvement of goiter induced by Propylthiouracil (Pro) in rats. Methods: The optimal modeling dose of Propylthiouracil was screened. After the models were established,SD female rats were randomly divided into the model group,8 μg/kg levothyroxine sodium group,Res groups (0. 2 g/kg、0. 4 g/kg) and the control group. All rats were administered with corresponding drugs or saline water for 6 weeks. Thyroid glands of rats were collected and weighed. The levels of serum free three iodine methyl adenine (FT3),free thyroxine (FT4),total T3,total T4 and thyroid stimulating hormone (TSH) were detected by biochemical analyzer. The thyroid pathological changes were observed by HE staining and Masson staining,the effect of Res on morphological indexes of thyroid gland were observed. Results: 5 mg/kg of propylthiouracil induced significant enlargement of thyroid gland in rats. Compared with the control group,the thyroid gland weight in the model group was significantly increased,the contents of FT3,FT4,T3 and T4 were decreased,TSH content was increased. The HE staining results showed that the thyroid follicular hyperplasia of rats in the model group was characterized by small follicular cavity and a large amount of glia in the follicular. The Masson staining results showed a large amount of collagen fiber hyperplasia in the model group. The rat HFE was significantly increased,while AFC was significantly decreased. Compared with the model group,the weight of thyroid gland was decreased after Res treatment,the contents of FT3,FT4,T3 and T4 were increased,the content of TSH was decreased. HE staining showed that the thyroid follicular was decreased,but the colloid in follicular was significantly increased.The Masson staining results showed that the collagen fibers in the model group significantly reduced,HFE was significantly increased while AFC was significantly decreased.
