摘要
目的:利用质谱流式技术建立研究肝细胞癌(HCC)肿瘤异质性的方法。方法:收集肝癌患者样本并制备成单细胞悬液。通过质谱流式技术用15种肿瘤干细胞标志物和28种免疫细胞相关的标志物对肝癌组织及其癌旁组织进行蛋白组学检测。结果:质谱流式分析有效地分出肝癌干细胞样亚群,并将免疫细胞分为25个细胞亚群。肝癌组织与其癌旁组织在肿瘤干细胞表达及免疫微环境组成方面存在差异。结论:质谱流式技术可实现对肿瘤干细胞样细胞和免疫细胞的精细分群,是检测肿瘤异质性的有效方法。
Objective:To establish the heterogeneity of hepatocellular carcinoma by mass spectrometry.Methods:We collected and isolated tissue from a case of liver cancer.Proteomics detection of hepatocarcinoma tissues was performed by mass spectrometry using 15 tumor stem cell markers and 28 immune cell-associated markers.Results:Mass spectrometry analysis effectively identified liver cancer stem cells-like subsets and divided the immune cells into 25 cell subpopulations.There was a difference in the expression of tumor stem cells and the composition of immune microenvironment between liver cancer tissues and its adjacent tissues.Conclusion:Mass spectrometry is an effective method to detect tumor heterogeneity by classify hepatocellular carcinoma stem cells and immune cells.
