摘要 为建立葛仙米中水溶性多糖含量的测定方法,对葛仙米水溶性多糖的预处理方法进行优化,并对苯酚-硫酸法测其多糖含量的方法学进行考察。结果表明:葛仙米水溶性多糖含量最佳的提取条件为水浴100℃、加热时间6 h、料液比1∶500,提取的多糖含量达32. 65%。葡萄糖标准溶液浓度(y)在0-0. 1 mg/mL范围内与吸光度值有较好的线性关系,线性方程y=0. 1202x-0. 0054,R2=0. 9997;苯酚-硫酸法测定葛仙米水溶性多糖的精密度、重现性和显色稳定性的相对标准偏差(RSD)分别为0、1. 289%和0. 584%,说明测定结果精密度高、重现性好、显色稳定;平均加样回收率为100. 13%、RSD为1. 18%,表明加样回收率高、结果可靠。 In order to establish a method for the determination of Nostoc sphaeroides kutz water-soluble polysaccharide content,the pretreatment method of the water-soluble polysaccharide were optimized and the methodology of the phenol-sulfuric acid method for determining the polysaccharide were inspected. Results: The optimal extraction conditions of Nostoc Sphaeroides kutz water-soluble polysaccharide were 100 ℃ of water bath,5 hours of heating time,1∶ 500 of liquid ratio. the content could reach up to 32. 18%. Glucose standard solution concentration( y) in the range of 0-0. 1 mg/mL and the absorbance value had good linear relationship,the linear equation was y = 0. 1202 x-0. 0054,R2= 0. 9997;The precision,reproducibility and color stability of the relative standard deviation( RSD) was 0,1. 289% and 0. 584%,indicating the determination results were high precision,good reproducibility and color stability;The sample recovery rate was 100. 13% and RSD was1. 18%,it showed that the sample recovery rate was high and the result was reliable.
