摘要
试验以前期分离筛选到的一株柑橘溃疡病生防内生细菌GN232为对象,采用单因素法优化其摇瓶发酵条件并测定其对脐橙苗期和成株期溃疡病的防治效果。结果表明,当菌株GN232采用YPD培养液、种子液按照5%量接种到100 mL、初始pH值为7的培养液中,在30℃、转数为200 r/min的振荡器中培养60 h后,获得的发酵液对柑橘溃疡病菌的抑制效果最强;内生细菌GN232在脐橙苗期和成株期树均表现出对溃疡病有一定的防治效果,其中当菌悬液稀释10倍时对脐橙苗期溃疡病的防效高于农用链霉素,菌悬液稀释50倍时对脐橙成年树溃疡病的防效能达到与农用链霉素相当的程度。试验结果为柑橘溃疡病的生物防治奠定了基础,亦可为生防内生细菌GN232的进一步研究和应用提供理论依据和技术支持。
In this experiment,an endophytic bacterium named GN232 with the abilty of controlling the citrus canker disease,which was obtained in the early stage,was taken as an object,and the shaking flask fermentation conditions were optimized by single factor method and its control effect on navel orange canker disease at the seedling stage and adult stage was determined.The results showed that when the seed solution of strain GN232 was inoculated into 100 mL culture medium with initial pH value of 7 in 5%quantity and the liquid was cultured in YPD medium with 200 r/min shaking frequency at 30℃ for 60 h,the fermentation broth had the strongest inhibitory effect on Xanthomonas citric subsp.citri.In the navel orange seedling and adult stages,endophytic bacterial strain GN232 had certain control effect on canker disease,and the control effect on navel orange seedling canker was higher than that of streptomycin when the suspension of endophytic bacteria GN232 was diluted 10 times,and the control effect of bacterial suspension on adult tree canker of navel orange was as good as that of streptomycin when diluted 50 times.The results laid a foundation for the biological control of citrus canker,and also provided theoretical basis and technical support for the further study and application of endophytic bacteria GN232.
