摘要
目的研究Circular RNAs(circRNAs)ZNF609在肺癌组织中的表达水平及其与肺癌患者临床病例特征、预后的关系,并探讨circZNF609在肺癌细胞中发挥的生物学功能。方法qRT-PCR检测circZNF609在50例肺癌组织和癌旁组织中的表达水平,并分析其在肺癌中的表达与肺癌患者临床病例特征、预后的关系;qRT-PCR检测circZNF609在肺癌细胞系中的表达;PC-9细胞经脂质体转染分为对照组(si-NC)和circZNF609 siRNA(si-circZNF609)组,MTS检测各组细胞增殖能力,流式细胞仪检测各组细胞周期阻滞情况,Transwell实验检测各组细胞转移能力,Boyden实验检测各组细胞侵袭能力。结果qRT-PCR结果显示circZNF609在肺癌组织中的表达显著高于癌旁组织,circZNF609的高表达与肺癌肿瘤分化及淋巴结转移显著相关,且circZNF609高表达的肺癌患者生存期较短(P值均<0.05)。circZNF609在肺癌细胞A549、PC-9、H226中的表达均显著高于人正常肺上皮细胞16HBE(P<0.05);转染circZNF609 siRNA后,PC-9细胞增殖、转移及侵袭能力均下降,G2期细胞增多(P值均<0.05)。结论circZNF609在肺癌组织和细胞系中均高表达,且高表达与肿瘤分化、淋巴结转移及预后不良相关,干扰circZNF609的表达显著抑制PC-9细胞的恶性生物学行为,circZNF609可能为肺癌的潜在治疗靶标。
Objective To investigate the expression of Circular RNAs(circRNAs)ZNF609 in lung cancer tissues and its relationship with clinical features and prognosis of lung cancer patients,and to explore the biological functions of circZNF609 in lung cancer cells.Methods qRT-PCR was used to detect the expression level of circZNF609 in 50 lung cancer tissues and adjacent tissues.The relationship between the expression of circZNF609 in lung cancer tissues and the clinical features and prognosis of patients was analyzed.qRT-PCR was used to detect the expression level of circZNF609 in lung cancer cells.PC-9 cells were transfected into the control group(si-NC)and the circZNF609 siRNA(si-circZNF609)group by lipofection,and MTS was used to detect cell proliferation ability.Flow cytometry cell was used to detect cycle arrest.Transwell assay was used to detect the cell metastasis ability.Boyden assay was used to detect the invasive ability of each group.Results qRT-PCR results showed that the expression of circZNF609 in lung cancer tissues was significantly higher than that in adjacent tissues.The high expression of circZNF609 was significantly associated with lung cancer tumor differentiation and lymph node metastasis,and lung cancer patients with high expression of circZNF609 had shorter survival(P<0.05).The expression of circZNF609 in lung cancer cells A549,PC-9 and H226 was significantly higher than that in human normal lung epithelial cells(P<0.05).After transfection of circZNF609 siRNA,the proliferation,metastasis and invasion ability of PC-9 cells were decreased.The number of cells in G2 cell cycle were increased(P<0.05).Conclusion circZNF609 is highly expressed in lung cancer tissues and cell lines,and its high expression is associated with tumor differentiation,lymph node metastasis and poor prognosis.Interfering with the expression of circZNF609 can significantly inhibit the malignant biological behavior of PC-9 cells,and circZNF609 may be a potential treatment target for lung cancer.
