摘要
硬实是种子常见的休眠类型之一,制约种子的种质创新利用。为研究硬实性的解剖结构,对不同种源的苦参种子的硬实率进行统计,利用石蜡切片法制作苦参种子及不同发育时期的种皮切片进行观察。结果表明,苦参种子具有很高的硬实率,高达93%;苦参种子由种皮、种脐及子叶等结构组成;种皮由角质层、栅栏组织、骨状石细胞、厚壁细胞以及薄壁细胞等组成,种皮结构较复杂,具有典型的豆科种皮结构特征;种脐部位由珠柄残余物和维管系统等组成;苦参子叶富含大量的淀粉粒。并且随着种子的发育,种皮的厚度不断增加,栅栏组织在种皮中所占的比例逐渐增加,这一特征可能是造成苦参硬实的原因之一。不同发育时期苦参种子解剖结构的观察结果可为进一步揭示苦参种子硬实性的形成机制奠定研究基础。
Hardseed is one of the common types of dormancy in seeds,which restricts the innovative use of seed germplasm.To study the hardseed anatomical structure,statistics on the hardness rate of the seeds of Sophora flavescens from different provenances,the seed slices of Sophora flavescens seeds and seed coats at different developmental stages were observed by paraffin sectioning.The results showed that Sophora flavescens seeds had a high hardseed rate,and the hardseed rate was 93%.The seeds of Sophora flavescens were composed of seed coat,umbilical cord and cotyledon,the seed coat consisted of stratum corneum,palisade tissue,bone stone cells,thick-walled cells and parenchyma cells.The seed coat structure was complex and had typical legume seed structure characteristics.The umbilical part was composed of the residue of the plant stem and the vascular system,the cotyledon of Sophora flavescens were rich in a large number of starch granules.And with the development of seeds,the thickness of the seed coat was increasing,and the proportion of palisade tissue in the seed coat was gradually increasing.This feature might be one of the reasons for the hardness.The observation of the anatomical structure of Sophora flavescens seeds in different developmental stages lays a foundation for further revealing the formation mechanism of the hard seed of Sophora flavescens seeds.
