石榴皮多组分提取工艺优化及多体系抗氧化活性评价 被引量:7

Extraction process optimization of pomegranate peel multi-component and evaluation on antioxidant activity in multi-system

摘要 目的:通过响应面法同时优化石榴皮中黄酮、多酚、皂苷的提取工艺,并多体系综合评价其抗氧化活性。方法:石榴皮进行超声辅助提取,以归一值(OD总值)作为复合指标对黄酮、多酚、皂苷进行响应面试验,并通过ABTS、DPPH、超氧阴离子、羟基自由基以及铅诱导的人胚肾细胞氧化应激通路对其进行体外抗氧化性考察。结果:优化得最优提取条件为乙醇浓度58%、液料比28∶1(mL/g)、提取时间53min、提取温度58℃。提取物对各自由基均具有较强的清除作用,且对铅诱导的Nrf2/Keap1/HO-1氧化应激通路具有抑制作用。结论:石榴皮多组分提取物具有清除自由基,减少氧化应激损伤的作用。 Objective: To optimize the extraction process of flavonoids, polyphenols and saponins in pomegranate peel by response surface method, and evaluate the antioxidant activity of pomegranate peel extract using mutli-system. Methods: Ultrasonic extraction of pomegranate peel was carried out and OD value was taken as composite index for flavonoids, polyphenols and saponins in response surface method, and investigated in vitro antioxidant activity by ABTS, DPPH, superoxide anion, hydroxyl radicals and lead-induced oxidative stress pathways in human embryonic renal cells. Results: The optimal extraction conditions was as follow: ethanol concentration was 58%, liquid-solid ratio was 28 : 1(mL/g), extracting time was 53 minutes, and extracting temperature was 58℃. The extract had superior eliminating effect on free-radicals and strong inhibitory effect on Nrf2/Keap1/HO-1 oxidative stress pathways. Conclusion: Pomegranate peel multi-component extract could eliminate free-radicals and reduce oxidative stress damage.
