响应面法优化女贞子的酒蒸工艺 被引量:4

Optimization of steaming with yellow rice wine process technology of Ligustri lucidi Fructus using response surface methodology

摘要 目的优化女贞子的最佳酒蒸工艺。方法采用高效液相色谱(HPLC)法测定酒蒸女贞子中红景天苷的含量,色谱柱为ZORBAX SB-C18(4.6 mm×250.0 mm,5.0μm),流动相为乙腈-0.1%磷酸水溶液,梯度洗脱,柱温25℃,检测波长224 nm,流速1 mL/min,进样量10μL。以女贞子中红景天苷的含量作为考察指标,采用单因素法考察黄酒用量、闷润时间以及蒸制时间对红景天苷含量的影响,并运用响应面法优化女贞子的酒蒸工艺。结果红景天苷的回归方程为Y=7 185.2X-40.251(r=0.999 5),表明红景天苷在0.020~0.600μg范围内具有良好的线性关系;精密度、重复性、稳定性均符合试验要求;红景天苷的平均回收率为100.71%,RSD为1.92%。响应面法优化的最优炮制工艺为:黄酒用量15.14%,闷润时间2.95 h,蒸制时间9.97 h。结论该法操作简便、准确,灵敏度、精密度高,分析速度快,可作为酒蒸女贞子药材的质量控制;响应面法优化女贞子酒蒸工艺方法简便可靠,能更好地保障酒女贞子的质量。 Objective To optimize steaming with yellow rice wine process technology of Ligustri lucidi Fructus.Methods The content of salidroside in yellow-rice-wine-steamed Ligustrum lucidum Fructus was determined by HPLC.The chromatographic column was ZORBAX SB-C18(4.6 mm×250.0 mm,5.0μm),acetonitrile-0.1%phosphoric acid aqueous solution as mobile phase,gradient elution,the detection wavelength was 224 nm and the flow rate was 1 mL/min,the column temperature was 25℃,and the sample size was 10μL.With the content of salidroside in Ligustrum lucidum Fructus as an index for investigation,the effects of the amount of yellow rice wine,the wetting time and the steaming time on the content of salidroside were investigated by single factor method,and steaming with the yellow rice wine process technology of Ligustrum lucidum Fructus was optimized by response surface methodology.Results The regression equation of salidroside was Y=7 185.2X-40.251(r=0.999 5),which showed that salidroside had a good linear relationship in the range of 0.020 to 0.600μg;the precision,repeatability and stability met the test requirements;the average recovery of salidroside was 100.71%,RSD was 1.92%.Response surface method was used to optimize the processing technology:the amount of yellow rice wine was 15.14%,the wetting time was 2.95 h,and the steaming time was 9.97 h.Conclusion The method is simple,accurate,sensitive and precise with quick analysis speed,and can be used for quality control of liquor-steamed Ligustrum lucidum Fructus.Response surface methodology is simple and reliable in optimizing yellow-rice-wine-steamed Ligustrum lucidum Fructus technology,which can better guarantee its quality.
