摘要
目的:探讨归芍地黄汤联合光动力疗法(Photodynamic Therapy,PDT)对大鼠脉络膜新生血管(Choroidal Neovascularization,CNV)中血管内皮细胞生长因子(Vascular Endothelial Growth Factor,VEGF)、碱性成纤维细胞生长因子(basic Fibroblast Growth Factor,bFGF)、色素上皮衍生因子(Pigment Epithelium Derived Factor,PEDF)表达干预的机理。方法:24只CNV模型大鼠随机平均分成阴性对照组、PDT组、中药组、中药联合PDT组,健康大鼠6只为正常对照组。各组大鼠做相应的中药和PDT治疗,每周行眼底荧光血管造影(Fluorescein Fundus Angiography,FFA)和光学相干断层成像(Optical Coherence Tomography,OCT)检查,5周后采用颈椎脱臼法处死大鼠,采用免疫组化检测视网膜VEGF、bFGF和PEDF积分光密度。结果:中药联合PDT组VEGF、bFGF积分光密度最低,PEDF积分光密度最高,VEGF/PEDF值下降最为显著,各组积分光密度比较差异有统计学意义(P<0.05)。结论:归芍地黄汤联合PDT可降低CNV大鼠模型VEGF、bFGF阳性表达,增强PEDF阳性表达,通过调节VEGF/PEDF值,促使CNV消退。
Objective:To explore the mechanism of the Guishao Dihuang decoction(归芍地黄汤)plus PDT on the expression of VEGF,bFGF and PEDF in choroidal neovascularization of rats.Methods:24 CNV model rats were randomly divided into negative control group,PDT group,TCM medicine group,TCM medicine plus PDT group.6 healthy rats were served as normal control group.Each group was treated with TCM medicine and PDT.FFA and OCT were performed weekly.After 5 weeks,the rats were killed by cervical dislocation.The integral optical density of retinal VEGF,B FGF and PEDF were detected by immunohistochemistry.Results:The integral optical density of VEGF and bFGF in the TCM medicine plus PDT group was the lowest,the integral optical density of PEDF was highest,and the value of VEGF/PEDF was lowest.The difference of integral optical density in each group was statistically significant(P<0.05).Conclusion:The Guishao Dihuang decoction plus PDT can reduce the positive expression of VEGF and bFGF in CNV rat model,enhance the positive expression of PEDF,and promote the regression of CNV by regulating the value of VEGF/PEDF.
