摘要
目的探讨水飞蓟素对多囊卵巢综合征(PCOS)模型大鼠炎症因子、纤维化指标及血糖相关指标、性激素的影响。方法将21日龄雌性SD大鼠随机分为正常组、模型组、二甲双胍组、水飞蓟素组及二甲双胍组+水飞蓟素组,每组10只。除正常组外,其余组大鼠均采用颈背部皮下注射脱氢表雄酮+注射用油剂方法造模,连续20 d。造模第21天,二甲双胍组给予二甲双胍灌胃,水飞蓟素组给予水飞蓟素灌胃,二甲双胍组+水飞蓟素组给予二甲双胍和水飞蓟素灌胃,正常组和模型组给予等量生理盐水灌胃,均1次/d,连续28 d。灌胃过程中,除正常组外,其余组均皮下注射脱氢表雄酮以防止其自身恢复排卵,并监测每组大鼠灌胃前及灌胃后1周末、2周末、3周末、4周末的体质量。实验结束后,对每组大鼠行腹腔葡萄糖耐量试验(IPGTT),测定空腹、2 h血糖及胰岛素水平;隔日下腔静脉取血测定血清睾酮(T)、卵泡刺激素(FSH)、黄体生成素(LH)、性激素结合球蛋白(SHBG)、白细胞介素-6(IL-6)、结缔组织生长因子(CTGF)水平。结果干预结束后,各组间体质量比较差异无统计学意义(P均>0.05),但各给药组体质量较模型组增加趋势减缓,二甲双胍组、水飞蓟素+二甲双胍组体质量增加趋势较水飞蓟素减缓更明显。模型组FINS、HOMA-IR、2hINS、2hPG、LH、T水平及LH/FSH比值均明显高于正常组(P均<0.05);二甲双胍组、水飞蓟素+二甲双胍组FINS、HOMA-IR、2hINS、2hPG及水飞蓟素组2hPG均明显低于模型组(P均<0.05);各给药组血清FSH、SHBG水平均明显高于模型组(P均<0.05),LH、T水平及LH/FSH比值均明显低于模型组(P均<0.05);水飞蓟素组与二甲双胍组各指标比较差异均无统计学意义(P均>0.05)。模型组血清IL-6、CTGF水平与正常组比较差异无统计学意义(P均>0.05);水飞蓟素组血清CTGF水平及水飞蓟素+二甲双胍组血清IL-6、CTGF水平均明显低于模型组(P均<0.0
Objective It is to investigate the effects of silymarin on inflammatory factors,fibrosis markers,blood glucose related indexes and sex hormones in rats with polycystic ovary syndrome(PCOS).Methods 21-day-old female Sprague-Dawley rats were randomly divided into normal group,model group,metformin group,silymarin group and metformin+silymarin group,with 10 rats in each group.Except the normal group,the rats in the other groups were treated with subcutaneous injection of dehydroepiandrosterone+injection oil for 20 days to establish the models.On the 21st day of modeling,metformin group was given metformin by gavage,silymarin group was given silymarin by gavage,metformin+silymarin group were given metformin and silymarin by gavage,and the normal group and model group were given the same amount of normal saline,1 time/d,for 28 consecutive days.During the gavage process,all the groups except the normal group were subcutaneously injected with dehydroepiandrosterone to prevent self-recovery of ovulation,and the body mass of the rats in each group was detected before gavage and at 1 weekend,2 weekends,3 weekends,4 weekends after gavage.At the end of the experiment,the peritoneal glucose tolerance test(IPGTT)was performed in each group of rats,and fasting,2h blood glucose and insulin levels were measured.Blood samples were taken from the inferior vena cava every other day to determine serum testosterone(T),follicle stimulating hormone(FSH),and luteinizing hormone(LH),sex hormone binding globulin(SHBG),interleukin-6(IL-6),and connective tissue growth factor(CTGF).Results After the intervention,there was no significant difference in body mass between every two groups(P>0.05),but the body weight of each drug group was increased slower than that of the model group,the body mass of metformin group,silymarin+metformin group was increased slower compared with silymarin group.The levels of FINS,HOMA-IR,2hINS,2hPG,LH,T and LH/FSH ratios in the model group were significantly higher than those in the normal group(P<0.05).The levels o
