荣筋拈痛方对骨关节炎作用靶点的分析及验证 被引量:10

Analysis and verification of the target of Rongjin Niantong Formula in the treatment of osteoarthritis

摘要 目的:BATMAN-TCM分析工具及R语言分析荣筋拈痛方对骨关节炎(OA)的作用及靶点并进行动物实验验证。方法:BATMAN-TCM分析工具预测荣筋拈痛方潜在作用靶点、发挥作用的方式途径及可能作用的疾病。R语言分析出OA模型大鼠与正常大鼠膝关节软骨组织的差异基因。韦恩图分析荣筋拈痛方潜在作用靶点与软骨组织差异基因的交集,选择靶点分子进行动物实验验证。将2月龄大鼠随机分为空白组、模型组、荣筋拈痛方组,空白组和模型组予0.9%氯化钠溶液灌胃,荣筋拈痛方组予荣筋拈痛方水提溶液灌胃。各组干预12周后,对关节软骨组织进行HE染色观察及Western Blot检测。结果:荣筋拈痛方具有632个化学成分,其中440个化学成分预测到1 787个潜在作用靶点,其通过多途径发挥作用,能治疗疼痛性疾病、心血管疾病、肾脏疾病、免疫性疾病、关节炎性疾病等多种疾病。OA大鼠软骨组织中差异表达的基因共226条。荣筋拈痛方潜在作用靶点与软骨组织差异基因相同的指标有34个,其中选择CXCR4进行验证。动物实验中HE染色及Western Blot检测显示,模型组软骨组织损伤且CXCR4蛋白表达升高(P<0.05),荣筋拈痛方组软骨组织损伤减轻且CXCR4蛋白表达降低(P<0.01)。结论:荣筋拈痛方能延缓OA软骨组织退变,作用机制与调节CXCR4表达有关。 Objective: To analyze the effects of Rongjin Niantong Formula on osteoarthritis(OA) by BATMANTCM analysis tool and the R programming language, and to preliminary verify the result by animal experiment. Methods: The BATMAN-TCM analysis tool was used to predict the targets and pathways in which Rongjin Niantong Formula may play a role, and diseases that may be treated by Rongjin Niantong Formula. The differential genes of knee joint cartilage tissue between OA model rats and normal rats was analyzed by the R programming language. The intersection of potential targets of Rongjin Niantong Formula and the differential genes of OA cartilage tissue were obtained in Venn diagram analysis. Targets were selected for subsequent experimental verification. Rats at 2 months age were randomly divided into the blank group, the model group and the Rongjin Niantong Formula group. The blank group and the model group were treated with normal saline, and the the Rongjin Niantong Formula group was treated with water extraction solution of Rongjin Niantong Formula. After the intervention, HE staining and Western Blot were performed on the articular cartilage tissue. Results: Rongjin Niantong Formula had 632 chemical components, of which 440 chemical components predicted 1 787 potential targets, which could treat a variety of diseases through multiple pathways, for example pain diseases, cardiovascular diseases, kidney diseases, immune diseases, arthritis diseases, and so on. The R programming language showed that a total of 226 genes were differentially expressed in cartilage tissue of OA rats.According to the Venn diagram analysis, there were 34 molecules in the intersection of potential targets of Rongjin Niantong Formula and the differential genes of OA cartilage tissue. CXCR4 was selected for verification. HE staining and Western Blot in animal experiments showed that the cartilage tissue of the model group was damaged and CXCR4 protein expression was increased(P<0.05).
