基于响应曲面优化的毛细管电泳测定通经草中的芦丁和异槲皮苷 被引量:2

Determination of Rutin and Quercetin-3-β-D-glucoside in Aleuritopteris Argentea by Capillary Electrophoresis Based on Optimization by Response Surface Method

摘要 目的采用高效毛细管电泳法(HPCE)分析通经草中的芦丁和异槲皮苷。方法采用响应曲面法(RSM)优化芦丁和异槲皮苷的毛细管电泳分离条件,建立HPCE测定通经草中芦丁和异槲皮苷含量的方法。结果在优化条件下,2种物质在10 min内得到良好分离。芦丁和异槲皮苷的峰面积和浓度分别在2~200,2~250μg/ml范围内呈良好线性关系。两者的检出限分别为0.6,0.4μg/ml。加标回收试验结果显示,芦丁和异槲皮苷平均回收率分别为107.9%,102.4%,RSD分别为4.3%,4.7%。结论该方法简便、快速、可靠,有望用于中草药通经草质量控制。 Objective To establish a high performance capillary electrophoresis(HPCE)method for determination of rutin and quercetin-3-β-D-glucoside in Aleuritopteris argentea.Methods Response surface method(RSM)was used to optimize the separation conditions of rutin and quercetin-3-β-D-glucoside by capillary electrophoresis,thus an HPCE method for the determination of rutin and quercetin-3-β-D-glucoside in Aleuritopteris argentea was established.Results Under optimum conditions,rutin and quercetin-3-β-D-glucoside were well separated within 10 min,and good linearity was found in the range of 2-200μg/ml and 2-250μg/ml for rutin and quercetin-3-β-D-glucoside,respectively.The detection limits of rutin and quercetin-3-β-D-glucoside were 0.6μg/ml and 0.4μg/ml,respectively.The average recoveries of standard addition of rutin and quercetin-3-β-D-glucoside were 107.9%and 102.4%,respectively,with RSDs of 4.3%and 4.7%,respectively.Conclusion The established method is simple,fast and reliable,and is hopeful to be used for quality control of Aleuritopteris argentea.
