摘要
自我国自然感染鸭乙型肝炎病毒(DHBV)的麻鸭肝脏,用蔗糖及氯化銫密度梯度离心纯化病毒核心颗粒(DHBcAg),其浮力密度范围为1.24~1.36g/ml,主要肽的分子量分别为32kD和18kD。用已知DHBV C基因编码的羧基端13个氨基酸合成肽制各能特异地与DHEcAg反应的抗体,并建立了检测DHBcAg的斑点酶免疫试验及ELISA法。初步用合成肽检测了感染鸭的抗-DHBc,发现在有DHEV复制的鸭血清中很少有抗-DHBc。
Core particles of duck hepatitis B virus( DHBV ) were purified from infected local duck liver tissues by sucrose and cesium chloride gradient ultracentrifugation.The buoyont density of these particles ranged 1.24-1.36g/ml and the molecular weight of the major peptides were 32 kD and 18 kD respectively. Based on the reported C gene sequence of DHBV, the carboxyl end 13 amino acid peptide was synthesized.Anti-serum raised versus this peptide could specifically react with DHBc Ag, and using this anti-serum, ELISA and dot-EIA were developed for assay of DHBcAg. Preliminary result for detection of anti-DHBc in infected ducks revealed that in ducks with active DHBV replication, only low titers of anti-DHBc could be detected.
