摘要
目的考察透骨消痛胶囊用药的安全性。方法 80只SD大鼠分4组,分别灌胃透骨消痛胶囊相当于成人用量的80、40、20倍和等体积生理盐水,4周后取一半大鼠进行血常规、生化、病理组织检测;免疫组化法观察细胞色素P450三种亚型在肝脏的表达;透射电镜观察肝、肾细胞超微结构。另一半大鼠停药1周,观察延迟毒性,指标同前。结果 1给药及停药后,大鼠一般状况、体重、血常规、生化指标及细胞色素P450三种亚型的表达与正常对照组无显著性差异;2病理组织、超微结构观察结果显示药物组与正常对照组无明显差异。结论透骨消痛胶囊安全性较好。
Objective To evaluate the safety of Tougu Xiaotong capsules(TGXT). Methods 80 SD rats were divided into 4 groups, including 3 experimental groups and a control group. The experimental groups were treated with different concentration of TGXT respectively, which were equivalent to 80, 40, 20 times of adult dosage, while the control group was given same volume of distilled water. After 4 weeks, half of the animals were killed to observe the histopathologic sections of organs, and the hematological and biochemical parameters were assessed. The expressions of CYP1A2, CYP2E1 and CYP3A4 were detected with immunohistochemistry. The cell ultrastructure of main organs was observed by transmission electron microscope.The remaining half of rats received stopping administration for 1 weeks, the delayed toxicity was observed and the above mentioned indexes were determined. Results All rats could tolerate the doses of TGXT. There was no significantly difference in general body conditions, body weight, hematological and biochemical parameters and expressions of CYP1A2, CYP2E1 and CYP3A4 among all the groups. There was also no significantly difference in changes of ultrastructure and pathohistology among all the groups. Conclusion TGXT has favourable safety.
