电喷雾差分电迁移率分析技术在生物研究中的应用 被引量:4

摘要 电喷雾—差分电迁移率分析(ES-DMA)可以快速获取1~500 nm范围的蛋白质或其他生物纳米粒子的粒径分布信息。它与电喷雾质谱技术的不同之处在于,质谱是根据质荷比区分,而ES-DMA分离主要基于粒子的大小和形状,不同大小、不同形状的粒子置于电场中时,将会以不同的速度发生迁移,通过测量气溶胶颗粒迁移的平均速率,最后即可转化得到粒子的粒径。ES-DM A技术作为新型的表征和定量技术手段,推动了生物纳米颗粒的定性和定量研究。总结了ES-DMA的基本原理及其在生物学研究领域中的应用,并对ES-DMA的发展趋势和应用前景作出了展望。 Electrospray–Differential Mobility Analysis(ES-DMA)is an emerging technique coupled with electrospray ionization and Differential Mobility Analysis.It is a type of ion mobility analyzer that could characterize the proteins and other bionanoparticles through rapidly obtained size distributions from1 nm to 500 nm.ES-DMA is different from electrospray ionization mass spectrometry which selects proteins based on m/z,while separation of proteins with ES-DMA is based on particles size and shape.When the particles with different sizes and shapes are placed in the electric field,they will migrate at different speeds.By measuring the average rate of the migration of aerosol particles,it can be converted to the size distributions of the particles.As a new technique of characterization and quantitation,ESDMA has greatly promoted the qualitative and quantitative research of biological nanoparticles.In this paper,we summarize the basic principles and application of ES-DMA in the field of biological research.Moreover,the trend of development and prospects of application of ES-DMA are also described.
