摘要
目的 研究三氧化二砷对猪髂动脉内皮细胞 (PIEC)的诱导凋亡作用。方法 采用MTT、细胞生长曲线、倒置相差显微镜、细胞爬片HE染色、细胞涂片苏丹Ⅲ染色、透射电镜等观察三氧化二砷作用后PIEC的变化。结果 PIEC细胞与三氧化二砷共孵育后出现细胞变圆、贴壁减少、上浮、细胞空泡样变性 ,脂肪颗粒增多 ;电镜等观察到核膜皱缩、核染色质凝结、边集等细胞凋亡现象 ,并且在一定的范围内该现象随药物浓度的增加而明显。结论 一定浓度的三氧化二砷能抑制PIEC细胞的生长 ,细胞脂肪变性 ,并诱导细胞凋亡。提示向肿瘤组织局部导入三氧化二砷可以抑制肿瘤局部供血动脉内皮的生长 ,促进供血动脉闭塞。
Purpose. To study the possible apoptosis-inducing effect of arsenic trioxide(As2O3) on pig iliac endothelium cell lines(PIEC). Methods. A wide range concentration of arsenic trioxide were cultured with PIEC with different time, cell growth curve, MTT assay, electron microscopy detection, inverted phase contrast microscope, HE staining, smear Sudan III staining etc. were used to study cell activity, morphologic changes. Results. After culturing with As2O3, PIEC present paste wall cells decrease, float, vacuolation, fat granule increase, and apoptotic features: i. e. cell membrane integrity, chromatin condensation, apoptotic body formation, etc. Conclusions. As2O3 can induce apoptosis of PIEC.
