摘要
目的建立一测多评法(QAMS)同时测定自制雷公藤提取物6种有效成分的含量。方法采用QAMS法,以雷公藤甲素、雷公藤内酯酮、雷酚内酯、雷公藤次碱、雷公藤红素和雷公藤内酯甲6种成分为研究对象,对外标法和校正因子法的实测值及计算值的差异进行分析。结果 6种指标成分在一定范围内线性关系良好,以雷公藤甲素为内参物,雷公藤内酯酮、雷酚内酯、雷公藤次碱、雷公藤红素和雷公藤内酯甲的fk/s分别为0.654、1.221、0.347、1.238、0.079,且在不同条件下重复性良好;3批样品中各成分含量分别为雷公藤甲素0.037~0.038mg/g、雷公藤内酯酮0.245~0.253 mg/g、雷酚内酯6.165~6.325 mg/g、雷公藤次碱14.439~15.065 mg/g、雷公藤红素1.699~1.856 mg/g、雷公藤内酯甲20.946~21.825 mg/g,且计算所得量与实测量测得结果无显著差异。结论QAMS法适用于雷公藤提取物的含量测定,可为雷公藤的质量控制提供参考。
Objective: To establish a multiple evaluation method(QAMS) to determine the content of 6 active ingredients of Tripterygium wilfordii extract including triptolide, triptonide, triptophenolide, wilfordine, celastrol and wilforlide A. Methods: 6 components were studied by QAMS method. The difference between the measured values and the calculated values of the external standard method and the correction factor method was analyzed. Results: The 6 indexes had good linear relationship in a certain range,and the fk/s of them were 0.654, 1.221, 0.347, 1.238, 0.079, respectively. The reproducibility was good under different conditions,and the contents of 3 batches of samples were 0.037~0.038 mg/g triptolide, 0.245~0.253 mg/g triptonide, 6.165~6.325 mg/g triptophenolide, 14.439~15.065 mg/g wilfordine, 1.699~1.856 mg/g celastrol and 20.946~21.825 mg/g wilforlide A, and there was no significant difference between the calculated amount and the measured results. Conclusion: A multiple evaluation method is applicable to the content determination of Tripterygium wilfordii extract, which can provide reference for the quality control of Tripterygium wilfordii.
