摘要
目的 观察艾滋病病毒外膜蛋白、核心蛋白与干扰素 (HIV -lenv/IFNα- 2b、HIV - 1gag/IFNα - 2b)融合基因的表达产物 ,能否作为理想的基因疫苗 ,使机体产生体液免疫和细胞免疫应答。方法 以小鼠为实验对象 ,用重组病毒vJ16env/IFNα - 2b、vJ38gag/IFNα - 2b免疫小鼠 ,以生理盐水和野生型痘苗病毒为对照组 ,检测小鼠外周血与脾淋巴细胞对ConA、Lps的反应性。用间接ELISA试验检测了重组病毒诱导小鼠产生的HIV -lenv/IFNα - 2b、HIV -lgag/IFNα - 2b抗体 ,用流式细胞仪测定小鼠脾细胞CD+ 4 、CD+ 8T细胞计数。结果 外周血与脾淋巴细胞对ConA、Lps的反应性 ,实验组与对照组比较差异有显著性意义 (P <0 0 5 )。vJ16env/IFNα - 2b与vJ38gag/IFNα - 2b能激发小鼠体内产生高滴度的抗体。CD+ 4 、T淋巴细胞与对照组比较差异有显著性意义 (P <0 0 5 )。CD+ 8T淋巴细胞与对照组比较无差异性 ,但呈增高趋势。结论 重组痘苗病毒能诱导小鼠产生较强的体液免疫和细胞免疫。重组痘苗病毒能在体外与HIV -lenv和HIV - 1gag阳性血清发生特异性反应 ,具有免疫原性和免疫反应性。
Objective To observe the acquired immunodeficiency syndrome virus exomembrane protein,core protein and Interferon (HIV-lenv/IFNα-2b,HIV-lgag/IFNα-2b) fusion genes expression product for gene vaccine,It induces humoral immunity and cellular immunity of organism.Methods The mice are immunized using the purified recombinant virus vJ16env/IFNα-2b,vJ38gag/IFNα-2b,as control with NaCl and vaccine.The ConA,Lps's response of T-lymphocytes in blood and spleen are examined by MTT\-\{570\}nm.OD value of HIV-lenv and HIV-lgag antibody is determined.CD + 4,CD + 8 T cell figure of mice spleen is examined by Flow Cytomeuy (FCM)Results There is significant change between test group and the control for ConA,Lps's response of T-lymphocytes in blood and spleen (P<0 05).vJl6env/IFNα-2b and vJ38gag/IFNα-2b could stimulate mice body to produce high level antibody ( P <0 05).CD + 4 T-lymphocytes significantly change between test group and the control( P <0.015).CD + 8T-lymphocytes do not significantly change between test group and the contral,but CD + 8 T-lymphocytes have the trend to increase.Conclusion Recombinant vaccinia virus could induce mice to producte humoral immunity and cellular immunity and could response HIV-lenv,HIV-lgag positive serum.It possesses immungenicity and immunereposibility.
