摘要 目的 探讨lncRNATMPO和miR-199a-5p调控胃癌SGC-7901细胞增殖和迁移的作用及其机制。方法采用qRT-PCR检测胃癌组织和细胞中lncRNATMPO-AS1的表达。采用CCK-8、Transwell检测胃癌细胞的增殖和迁移;采用生物信息分析、荧光素酶实验、qRT-PCR验证lncRNATMPO-AS1和miR-199a-5p的靶向关系。结果 与癌旁正常组织和正常胃黏膜上皮细胞相比,lncRNATMPO-AS1在胃癌组织和细胞中表达上调(P<0.05)。下调lncRNATMPO-AS1增加胃癌细胞的增殖、迁移和侵袭能力。lncRNATMPO-AS1靶向作用miR-199a-5p并下调其表达水平。结论lncRNATMPO-AS1在胃癌中发挥促癌效应,其可以通过抑制miR-199a-5p的功能而促进胃癌细胞的增殖和迁移。 Objective To investigate the effect and mechanism of long chain non-coding RNA TMPO-AS1 and miR- 199a-5p on proliferation and migration of gastric cancer SGC- 7901 cells. Methods The expressions of lncRNA TMPO-AS1 was detected by real time qRT-PCR.,and the proliferation and migration of gastric cancer SGC-7901 cells were detected by CCK-8 and transwell,and the targeting relationship between lncRNA TMPO-AS1 and miR-199a-5p was verified by luciferase assay,Bioinformatics analysis and qRT-PCR. Results The expression of lncRNA TMPO-AS1 was up-regulated in gastric cancer tissues and cells. Down-regulation of lncRNA TMPO-AS1 increased the proliferation and migration of gastric cancer cells. Bioinformation analysis,luciferase assay. lncRNA TMPO- AS1 targeted miR-199a-5p and down-regulated its expression level. The results further validated that miR-199a-5p could reverse the promotion of lncRNA TMPO-AS1 on gastric cancer cells. Conclusion lncRNA TMPO-AS1 could promote the proliferation and migration of gastric cancer cells by inhibiting the function of miR-199a-5p.
