摘要
采用电镜研究花瓣的形态学特征,是对花卉花瓣进行系统进化关系研究和品种鉴定的重要手段之一,可以为花卉新品种的选育,以及为切花和花卉保鲜技术提供微观形态依据。由于花瓣的制样过程直接影响对其形态观察的电镜结果,本研究通过控制变量法,对戊二醛固定液的浓度、花瓣固定时间以及脱水干燥方法等条件进行考察,最终通过电镜扫描观察结果确定了最优的花瓣制样方法。结果显示:花瓣在浓度为7.5%的戊二醛溶液中固定4 h,用梯度无水乙醇进行逐级脱水,再用梯度叔丁醇逐级置换,以及冷冻干燥,经过电镜观察验证,得到表面结构清晰的月季花瓣扫描电镜图片。
The use of electron microscopy has showed its importance in study of morphological characteristics of flower petal, such as in phylogenetic relationship research and variety identification of flowers. It provides microscopic morphology basis for the varieties breeding and for cut-flower and flower preservation techniques. The petal preparation process directly affects the effect of the morphological observation by electron microscopy. In this study, different processing conditions, sucn as the concentration of glutaraldehyde fixative, the time of petal fixation and the process of dehydration and drying, were investigated by controlled variable method. Finally, the optimal method of petal preparation was determined by scanning electron microscopy. The results showed that the microscopic morphological structure can be clearly observed by scanning electron microscopy image, when the petals were fixed in 7.5 % glutaraldehyde solution for 4 h, dehydrated with gradient absolute ethanol, then replaced with gradient t-butanol, and freeze-dried.
