GNPS指导下濒危药材重楼微生物次生代谢产物的研究 被引量:4

摘要 目的研究濒危药材重楼微生物及其次生代谢产物的指纹图谱和GNPS网络,以发现其与宿主相同或相似的化学成分。方法采用不同培养基分别从重楼根、茎、叶、根状茎及根系土壤中得到重楼相关微生物;采用ITS及D1/D2方法对菌株进行种属鉴定;采用TLC、LC-MS/MS等技术建立指纹图谱和GNPS网络,并对其化学成分进行初步分析。结果从重楼不同部位共分离得到12株真菌、16株细菌和4株放线菌;其中14株微生物种属鉴定结果表明共涉及9个菌属;次生代谢产物的类型共涉及螺甾烷醇类、偏诺皂苷元类、苯丙素类、黄酮类、蒽醌类及甾体皂苷等化合物;LC-MS和GNPS技术共检测出15个与宿主成分相同或类似的化合物。结论重楼含有丰富的内生及根际相关微生物,首次采用GNPS技术对其次生代谢产物进行分析,并得到与宿主相同或相似的化学成分,为后续重楼微生物次生代谢产物的分离和新颖骨架化合物的发现提供前期基础。 Objective To study the fingerprint of the secondary metabolites from endangered Chinese medicinal materials Paris polyphylla,endophytic and Global Natural Products Social network(GNPS),in order to find the same or similar chemical constituents as the host.Methods The endophytic and environmental microorganisms from roots,stems,leaves,rhizomes and soils of Paris polyphylla based on the different media were isolated and purified.The name of microorganism was subjected by ITS or D1/D2.The fingerprints and GNPS were established by TLC and LC-MS/MS,and the secondary metabolites of microorganism were analyzed.Results 12 fungi,16 bacteria and 4 actinomyces were isolated from Paris polyphylla.14 of the fungi and bacteria belonged to nine genus,and secondary metabolites from them included spirosterols,penonosaponins,phenylpropanoids,flavonoids,anthraquinones and steroidal saponins,etc.15 compounds were found as the same as or similar with secondary metabolites from the Paris polyphylla according to GNPS.Conclusion There are abundant endophytic and environmental microorganisms in the Paris polyphylla.The GNPS is used to analyze the secondary metabolites from Endangered Paris polyphylla-Associated microorganisms for the first time,and the same or similar chemical constituents as the host have been obtained,which provides a preliminary basis for the subsequent isolation of secondary metabolites from microorganisms and the discovery of novel skeleton compounds.
