摘要
目的观察依达拉奉腹腔注射对大鼠缺血性脑损伤的治疗作用,并探讨其机制。方法将36只SD大鼠随机分为3组各12只,Edv组、MCAO组均采用大脑中动脉闭塞(MCAO)方法制备缺血性脑损伤模型,Sham组仅分离暴露血管不做栓塞;Edv组于MCAO术后1h腹腔注射8mg/kg依达拉奉,连续3 d;Sham组及MCAO组分别在同时间点腹腔注射同量生理盐水。于MCAO术后24h行神经功能缺损评分;于MCAO术后5天采用氯化三苯四氮唑(TTC)染色法测算大鼠脑梗死体积;采用Western blotting法检测脑组织Notch通路相关蛋白Notch-l、NICD、 HeS-l和小胶质细胞标志物Iba-1蛋白;采用ELISA法检测脑组织IL-6、IL-1β、TNF-α。结果与Sham组比较,Edv组、 MCAO组神经功能缺损评分和脑梗死体积百分比增加(P均<0. 05);与MCAO组比较,Edv组神经功能缺损评分和脑梗死体积百分比减少(P均<0. 05)。与Sham组比较,Edv组、MCAO组Notch-l、NICD、HeS-l、Iba-l蛋白表达升高(P均<0. 05);与 MCAO 组比较,Edv 组 Notdi-1、NICD、Hes-1、Iba-1蛋白表达降低(P均<0. 05)。与 Sliam 组比较,Edv组、MCAO组 IL-6、IL-1β、TNF-α水平升高(P均<0.05);与 MCAO 组比较,Edv 组 IL-6、IL-1β、TNF-α水平降低(P均<0.05)。结论依达拉奉对大鼠缺血性脑损伤有治疗作用,其可有效改善缺血性脑损伤大鼠的神经功能症状、减少脑梗死体积,机制可能是通过下调Notch信号通路,抑制小胶质细胞活化,减少炎症因子释放而实现。
Objective To observe the therapeutic effect of intraperitoneal injection of edaravone on ischemic brain in- juiy in rats and to explore its mechanism. Methods Thirty-six SD rats were randomly divided into 3 groups with 12 rats in each group. Middle cerebral arteiy occlusion (MCAO) method was used to prepare ischemic brain injuiy models in the Edv group and MCAO group, while we only isolated and exposed the blood vessels without embolization in rats of the Sham group. Rats in the Edv group were intraperitoneally injected with 8 mg/kg edaravone 1 h after MCAO for 3 consecutive days, while Sham group and MCAO group with the same amount of normal saline at the same time. Nerve function defect score was performed at 24 h after MCAO. The volume of cerebral infarction in rats was detected by TTC staining 5 days af-ter MCAO. The Notch pathway-related proteins Notch-1, NICD, Hes-1, and microglial marker Iba-1 in the brain tissues were detected by Western blotting. IL-6, IL-1β and TNF-α in the brain tissues were detected by ELISA. Results Com-pared with the Sham group, Edv group and MCAO group showed increased neurological deficit score and cerebral infarction volume percentage (both P < 0. 05). Compared with the MCAO group, Edv group showed decreased neurological deficit score and cerebral infarction volume percentage (both P <0. 05). Compared with the Sham group, the protein expression levels of Notch-1,NICD,Hes-1,and Iba-1 increased in the Edv group and MCAO group (all P <0. 05). Compared with the MCAO group, Edv group showed decreased protein expression levels of Notch-1 , NICD,Hes-1,and Iba-1 (all P< 0. 05). Compared with the Sham group, the IL-6, IL-1β, and TNF-α levels increased in the Edv group and MCAO group (all P < 0. 05 ). Compared with the MCAO group, the levels of IL-6, IL-1β and TNF-α in the Edv group decreased (all P<0.05).
