自噬相关蛋白在肝泡型包虫病中的表达及意义

Expression and significance of autophagy-associated protein in hepatic alveolar echinococcosis

摘要 目的探讨哺乳动物雷帕霉素靶蛋白(mammalian rapamycin,mTOR)、微管相关蛋白轻链3蛋白(microtubule-associated protein 1 light chain 3,LC3)及bcl-2同源结构域蛋白(bcl-2 homeodomainprotein,Beclin-1)等自噬相关蛋白在肝泡型包虫病组织中的表达水平,初步分析自噬与肝泡型包虫病之间的关系。方法收集肝泡型包虫病患者术后正常肝组织及边缘带样本,用免疫组织化学方法检测LC3、Beclin-1、mTOR在肝泡型包虫病边缘组织及正常肝组织的表达水平;用Tissue FAXS PLUS定量系统分析各个蛋白的表达强度。结果LC3、Beclin-1在泡型包虫病灶边缘组织表达水平高于正常肝组织(P<0.05);mTOR在泡型包虫病灶边缘肝组织中表达水平低于正常肝组织(P<0.001)。结论在肝泡型包虫病进展过程中,自噬可能参与其中。 Objective To investigate the expression level and significance of autophagy-related protein(mammalian rapamycin(mTOR),microtubule-associated protein 1 light chain 3(LC3)and bcl-2 homeodomain protein(Beclin-1))in hepatic alveolar echinococcosis and preliminarily analyzed the association between autophagy and hepatic alveolar echinococcosis.Methods The normal liver tissue and lesion edge tissue were collected in patients with hepatic alveolar echinococcosis.The immunohistochemistry method was used to assess the expression levels of LC3,Beclin-1 and mTOR.The Tissue FAXS PLUS quantitative system analyzed the expression intensity.Results The expression levels of LC3 and Beclin-1 in lesion edge tissue were higher than normal liver tissues(P<0.05).The expression level of mTOR in lesion edge tissue was lower than normal liver tissue(P<0.001).Conclusion Autophagy maybe involved in the progression of hepatic echinococcosis.
