摘要
目的:探究急性分泌性中耳炎(SOM)患者耳积液白介素2(IL-2)、白介素6(IL-6)与外周血辅助细胞/杀伤抑制细胞(CD4^+/CD8^+)的相关性.方法:选取2015年1月~2016年12月我院确诊为急性分泌性中耳炎的51例患者作为观察组,选取同一时段我院体检的52例健康人群作为对照组.应用酶联免疫吸附法(ELISA)检测IL-2、IL-6水平,采用美国贝克尔曼库尔特公司FC500液式细胞仪检测CD4^+、CD8^+.比较两组研究对象CD4^+、CD8^+及CD4^+/CD8^+比值结果及两组IL-2、IL-6水平,并分析IL-2、IL-6与外周血CD4^+/CD8^+的相关性.结果:观察组中CD4^+、CD8^+及CD4^+/CD8^+比值均高于对照组,两组之间差异均具有统计学意义.比较观察组与对照组外周血中IL-2、IL-6水平显示,二者差异不具有统计学意义;但观察组外周血及对照组外周血中IL-2、IL-6水平均低于观察组中耳积液水平,其差异均具有统计学意义.采用Pearson对IL-2、IL-6与外周血CD4^+/CD8^+的相关性进行分析,结果显示,IL-2、IL-6与外周血CD4^+/CD8^+呈显著正相关.结论:IL-2、IL-6水平与CD4^+、CD8^+及CD4^+/CD8^+比值的高表达可作为急性分泌性中耳炎患者病情变化的预测指标,且IL-2、IL-6与CD4^+/CD8^+呈显著正相关,可为临床研究提供参考.
Objective To investigate Correlation between interleukin-2(IL-2), interleukin-6(IL-6) in ear effusion and peripheral blood helper/suppressor(CD4^+/CD8^+) in peripheral blood of patients with acute secretory otitis media(SOM). Methods 51 cases of acute SOM(observation group) and 52 cases of health examinees(control group) in our hospital from January 2015 to December 2016 were selected. The levels of inflammatory factor(IL-2, IL-6) and Tregs cell(CD4^+, CD8^+) were detected by enzyme-linked immunosorbent assay(ELISA) and FC500 flow cytometry(Beckman Coulter, USA), respectively. Then the correlation was analyzed. Results The CD4^+, CD8^+ and CD4^+/CD8^+ in the observation group were higher than those in the control group, while no statistic difference was found between groups in the IL-2 and IL-6 levels. However, the IL-2 and IL-6 levels in the peripheral blood of both groups were lower than those in the ear effusion. Pearson correlation analysis showed that IL-2 and IL-6 levels were positively correlated with CD4^+/CD8^+ in peripheral blood. Conclusion The levels of IL-2, IL-6, CD4^+, CD8^+ and CD4^+/CD8^+ can be used to predict the severity of the acute SOM, and there is a positive correlation between IL-2, IL-6 and CD4^+/CD8^+, which can provide a reference for the clinical study.
