冰醋酸对水溶性伊红Y染液的影响及应用比例 被引量:1

摘要 目的探讨水溶性伊红Y染液中未加入冰醋酸时和逐步加入冰醋酸后其染色能力的变化及在实际应用中应加入的合适比例使染液能达到最佳的染色效果。方法选取近期接收的标本,常规脱水包埋切片。在各组切片进行染色时,在水溶性伊红Y染液内逐步加入冰醋酸,并记录下各组切片染色时水溶性伊红Y染液内的冰醋酸的量,最后显微镜下观察各组切片的染色效果以明确目的。结果当水溶性伊红Y染液内未加入冰醋酸时其染色能力较差,切片不易染色。随着染液内加入的冰醋酸逐渐增多,染液的染色能力逐渐增强,切片内各物质染色越深。当染液内加入的冰醋酸比例约0.5%时,染液的染色效果最佳,细胞核与细胞浆的对比度较好,可符合诊断要求。加入过多冰醋酸后染液的染色能力过强,切片被伊红深染,细胞核也被染色而呈现紫色,对比度下降,从而影响诊断。结论冰醋酸可使水溶性伊红Y染液的染色能力增强。但加入过多的冰醋酸后水溶性伊红Y染液的染色能力过强,切片的整体染色效果反而下降。因此在实际应用中冰醋酸的量需控制在一定范围(约0.5%时)内,使染液的染色能力适中,使细胞质易被伊红染液染成不同色调的红色而细胞核不被染色,从而能形成鲜明对比,达到诊断要求。 Objective To explore the dyeing ability of water-soluble eosin Y dyeing solution without adding glacial acetic acid and after gradually adding glacial acetic acid,and to explore the appropriate ratio should be added in practical application to achieve the best dyeing effect.Methods The recently received specimens were selected and the conventional dehydrated embedded sections were used.When staining each group of slices,the amount of glacial acetic acid was gradually added to the water-soluble eosin Y dye solution,and the amount of glacial acetic acid in the water-soluble eosin Y dyeing solution was recorded under the staining each group of slices.The staining effect of each group of slices was used for a clear purpose.Results When glacial acetic acid was not added to the water-soluble eosin Y dyeing solution,its dyeing ability was poor,and the slice was not easy to stain.As the glacial acetic acid added to the dyeing solution gradually increased,the dyeing ability of the dyeing solution gradually increased,and the dyeing of the substances in the slices was deeper.When the proportion of glacial acetic acid added to the dyeing solution was about 0.5%,the dyeing effect of the dyeing solution was the best,and the contrast between the nucleus and the cytoplasm was good,which could meet the diagnostic requirements.After adding too much glacial acetic acid,the staining ability of the dyeing solution was too strong,the slice was deeply stained with eosin,the nucleus was also stained to appear purple,and the contrast was decreased,thereby affecting the diagnosis.Conclusion Glacial acetic acid can enhance the dyeing ability of water-soluble eosin Y dyeing solution.However,the dyeing ability of the water-soluble eosin Y dye solution is too strong after adding too much glacial acetic acid,and the overall staining effect of the slice decreases.Therefore,in practical applications,the amount of glacial acetic acid needs to be controlled within a certain range(about 0.5%),so that the dyeing ability of the dyeing solution is mod
