老龙七中苔色酸乙酯的分离及体外抗菌活性研究 被引量:1

摘要 目的 对中药老龙七中苔色酸乙酯进行提取分离,并考察苔色酸乙酯对大肠杆菌和金黄色葡萄球菌的体外抗菌活性。方法 采用硅胶柱层析、Sephadex LH-20葡聚糖柱层析和重结晶技术分离老龙七中的苔色酸乙酯,并通过核磁共振波谱和质谱确定其结构;以硅胶薄层层析法和高效液相色谱法确定化合物的纯度;以试管倍比稀释法测定苔色酸乙酯对大肠杆菌和金黄色葡萄球菌最低抑菌浓度(MIC)和最小杀菌浓度(MBC);采用菌落计数法绘制2种菌株的时间-杀菌曲线(KCS)。结果 从老龙七提取液的氯仿萃取相中分离得到了1个化合物,经光谱数据确定其结构为苔色酸乙酯;硅胶薄层层析检测在三种不同展开剂系统中展开均为单一斑点,以高效液相色谱法检测仅出现单一峰;该化合物对大肠杆菌的MIC值为256μg·mL^-1,MBC值为1024μg·mL^-1,对金黄色葡萄球菌的MBC值为128μg·mL^-1;通过菌落计数法绘制出2种菌株的时间-杀菌曲线(KCS)。结论 中药老龙七植物中存在较多量的苔色酸乙酯,其对大肠杆菌、金黄色葡萄球菌具有一定的体外抗菌效果。 OBJECTIVE To isolate and determine the compound from the lichenplant Lobaria kurokawae, and to investigate its antibacterial activities against Escherchia coli and Staphylococcus aureus in vitro.METHODS One compound was obtained by using various chromatographic techniques and its str ucture was determined on the basis of spectral data (MS,and 1H-NMR).The constituent was detected by thin layer chromatography and its purity was confirmed by high-performance liquid chromatography (HPLC).Minimum inhibitory concentrations (MIC) and minimum bactericidal concentration (MBC) to two standard bacteria strains were tested by double dilution method.Time-kill curve was drew according to colony count. RESULTS One compound was isolated and was determined to be ethyl orsellinate.Its MIC value to Escherchia coli and Staphylococcus aureus was 0.256μg·μL^-1 and 0.128μg·μL^-1 respectively .The MBC value to Escherchia coli was 1.024μg·mL^-1 .CONCLUSION The lichen plant contained ethyl orsellinate and the constituent has antibacterial activity to Escherchia Coli and Staphylococcus aureus in vitro.
