摘要
目的:探讨卡那单抗对慢性荨麻疹小鼠外周血1型辅助性T细胞/2型辅助性T细胞(Th1/Th2)平衡及相关细胞因子表达水平的影响。方法:将50只昆明成年小鼠(雌雄各半)随机分为正常对照组、模型组、卡那单抗低剂量、中剂量和高剂量组。模型组和卡那单抗各剂量组小鼠经腹壁皮内注射抗血清建立慢性荨麻疹模型,同时卡那单抗各剂量组皮下注射相应药物剂量,正常对照组和模型组皮下注射等量生理盐水,2周后进行抗原攻击。取各组小鼠眼眶血分离血清,分别采用流式细胞仪检测和酶联免疫吸附试验(ELISA)法测定血清Th1、Th2表达水平和白细胞介素(IL)-2、IL-4、IL-6、IL-10、肿瘤坏死因子-α(TNF-α)和γ干扰素(IFN-γ)的含量。结果:与正常对照组比较,模型组和卡那单抗各剂量组小鼠血清Th1表达水平及IL-2、IL-4、IL-6、TNF-ɑ和IFN-γ含量升高,Th2表达水平降低(均P<0.05)。与模型组比较,卡那单抗中、高剂量组Th1表达水平及IL-2、TNF-α和IFN-γ含量下降,Th2表达水平及IL-6和IL-4含量升高(均P<0.05)。与卡那单抗中剂量组比较,卡那单抗高剂量组Th1表达水平及IL-2、TNF-α和IFN-γ含量下降,Th2表达水平及IL-6、IL-4和IL-10含量升高(均P<0.05)。结论:卡那单抗可以通过调节炎症因子抑制炎性细胞活性改善慢性荨麻疹小鼠外周血Th1/Th2平衡,对治疗慢性荨麻疹可能有一定的疗效。
Objective:To investigate the effect of Canakinumab on the balance of type 1 helper T cell/type 2 helper T cell(Th1/Th2)and the expression of related cytokines in peripheral blood of mice with chronic urticaria.Methods:The total of 50 Kunming adult mice(half male and half female)were randomly divided into normal control group,model group,low dose group,middle dose group and high dose group.Mice in the model group and various doses of Canakinumab groups were injected with anti-serum through abdominal wall to establish the model of chronic urticaria,and mice in the different doses of Canakinumab were subcutaneously injected with corresponding drug dose,normal control group and model group mice were subcutaneously injected with the same amount of normal saline,and antigen attack was conducted 2 weeks later.The expression levels of Th1 and Th2 were determined by flow cytometry,and the levels of IL-2,IL-4,IL-6,IL-10,TNF-αand IFN-γin serum were detected by enzyme-linked immunosorbent assay(ELISA).Results:Compared with the normal control group,the expression of Th1 and the contents of IL-2,IL-4,IL-6,TNF-αand IFN-γwere increased,while the expression of Th2 was decreased in the model group and Canakinumab group(P<0.05).Compared with the model group,the Th1 expression and the IL-2,TNF-ɑand IFN-γcontents decreased while the Th2 expression and the concentrations of IL-6 and IL-4 increased in the middle and high dose groups,and the changes were more marked in high dose group(P<0.05).Conclusion:Canakinumab could improve the balance of Th1/Th2 in peripheral blood of chronic urticaria mice by regulating inflammatory factors to inhibit the activity of inflammatory cells.
