圆珠半夏带顶芽块茎组培快繁体系的构建 被引量:1

摘要 为半夏种苗繁育提供参考,采用组织培养法,通过单因素试验和3因素4水平正交试验,探索6-BA、NAA、2,4-D生长调节剂不同浓度及其组合对圆珠半夏顶芽萌发、丛生芽诱导和愈伤组织诱导的影响。结果表明:不同浓度6-BA、NAA、2,4-D及其组合对顶芽萌发、愈伤组织和丛生芽诱导具有显著或极显著促进作用。单因素试验中,1.0mg/L6-BA、0.2mg/LNAA、0.2mg/L2,4-D诱导的愈伤组织块数最多,分别为26块、29块和24块。以圆珠半夏带顶芽块茎为外植体进行组织培养时,愈伤组织形成的最优组合为MS+6-BA1.0mg/L+NAA0.3mg/L+2,4-D0.3mg/L,诱导顶芽萌发和丛生芽形成的最优组合为MS+6-BA1.2mg/L+NAA0.3mg/L+2,4-D0.2mg/L。 To provide references for Pinellia ternata seedling breeding,the effects of 6-BA,NAA and 2,4-D on apical bud germination and cluster bud induction were studied by means of tissue culture and orthogonal experiment with three factors and four levels.Results:Different concentrations of 6-BA,NAA,2,4-D and their combinations had significant or extremely significant effects on apical bud germination,callus induction and cluster bud induction.In single factor experiment,callus blocks induced by 1.0 mg/L 6-BA,0.2 mg/L NAA,0.2 mg/L 2,4-D were the most,which were 26,29 and 24,respectively.When the tubers of P.ternata with apical buds were used as explants for tissue culture,the optimal combination for callus induction was MS+6-BA 1.0 mg/L+NAA 0.3 mg/L+2,4-D 0.3 mg/L.The optimum combination of inducing apical bud germination and cluster bud formation was MS+6-BA 1.2 mg/L+NAA 0.3 mg/L+2,4-D 0.2 mg/L.
