慢性失眠患者血清胶质细胞损害标志物研究 被引量:2

A study on markers of glial cell damage in patients with chronic insomnia

摘要 目的探索慢性失眠(chronic insomnia,CI)患者血清S100钙结合蛋白B(S100 calcium binding protein B, S100B)和胶质纤维酸性蛋白(glial fibrillary acidic protein,GFAP)水平变化及其与睡眠质量和认知功能间关系。方法纳入CI患者60例,以32名健康体检者为对照。用匹兹堡睡眠质量指数(Pittsburgh sleep quality index,PSQI)评估失眠程度,蒙特利尔认知评估量表(Montreal cognitive assessment,MoCA)评估总体认知功能,九盒迷宫评估特异性记忆功能,ELISA检测血清S100B和GFAP含量。结果与对照组比较,CI患者PSQI分高(t=28.591, P<0.001),MoCA分低(Z=-3.645, P<0.001),物体工作(Z=2.973, P=0.003)、空间工作(Z=-5.166, P<0.001)和物体再认(Z=-2.613, P=0.009)记忆的错误数均较多,血清S100B和GFAP增高(t=5.574, P<0.001;t=-4.150,P<0.001)。S100B与PSQI分正相关(r=0.311, P=0.035),与MoCA分负相关(r=-0.320, P=0.030);GFAP与空间工作(r=0.346,P=0.011)、物体再认记忆错误数(r=0.277, P=0.043)正相关。结论 CI患者血清S100B和GFAP水平升高,并与睡眠质量和认知功能损害相关。 Objective To explore whether serum levels of S100 calcium binding protein B (S100B) and glial fibrillary acidic protein (GFAP) change, and these changes, if it is case, are correlated with sleep quality and cognitive function in the patients with chronic insomnia (CI). Methods Sixty CI patients and thirty-two healthy controls were enrolled. Pittsburgh Sleep Quality Index (PSQI), Montreal Cognitive Assessment scale (MoCA) and Nine-Box Maze Test were used to assess the insomnia severity, general cognition and specific memories, respectively. The serum levels of S100B and GFAP were detected using ELISA. Results Compared to the controls, CI patients had significantly higher PSQI score (t=28.591, P<0.001) and lower MoCA score (Z=-3.645, P<0.001). The CI patients had significantly higher errors in object working (Z=-2.973, P=0.003), spatial working (Z=-5.166, P<0.001) and object recognition (Z=-2.613, P=0.009) memories in the Nine-Box Maze than the controls, with higher S100B and GFAP levels (P<0.001). The Partial correlation analysis showed that in CI patients, the levels of S100B and GFAP correlated with PSQI score positively (r=0.311, P=0.035;r=0.361, P=0.006) and MoCA score negatively (r=-0.320, P=0.030;r=0.356, P=0.007). The GFAP level also positively correlated with the errors in spatial working (r=0.346, P=0.011) and object recognition (r=0.277, P=0.043) memories. Conclusion The levels of S100B and GFAP is increased in CI patients, which, specifically GFAP is correlated with insomnia severity and cognitive impairment.
