摘要
为确定花生苗期耐盐鉴定的适宜NaCl浓度和评价指标,本试验以4个耐盐性不同的花生品种白沙1016、丰花2号、山花18号和ICGV86699为材料,将种子置于生长室进行萌发,后利用水培法培养8 d再进行不同浓度NaCl处理(100、200、300 mmol/L),以Hogland营养液培养的为对照,观察、记录盐处理下不同花生植株的萎焉程度,测定主茎高、茎叶鲜重、茎叶干重、根长、根鲜重、根干重、根表面积和根体积等性状指标,并对不同品种的植株性状相对值,及其与耐盐指数的相关性进行分析。结果表明:水培法鉴定花生苗期耐盐性适宜的NaCl浓度为200 mmol/L,植株性状测定适宜时间为NaCl胁迫处理7 d;丰花2号、白沙1016、山花18号和ICGV86699在200 mmol/L NaCl胁迫处理7 d时,耐盐系数分别是0.77、0.72、0.64和0.53,耐盐指数为0.85、0.89、0.60、0.39,品种间耐盐性差异显著。相关性分析显示,耐盐系数、相对茎叶干重、相对根长与耐盐指数呈极显著正相关,相关系数分别为0.95、0.95、0.90,因此,耐盐系数、相对茎叶干重、相对根长可作为花生苗期耐盐鉴定的评价指标。
In order to determine the suitable NaCl concentration and evaluation indicators for salt tolerance identification at peanut seedling stage, four peanut varieties with different salt tolerance, Baisha 1016, Fenghua 2, Shanhua 18 and ICGV86699, were used as materials. The seeds were germinated in the growth chamber, then cultured in water for 8 days and treated with NaCl at different concentrations(100, 200, 300 mmol/L). The wilting degree of peanut plants under salt treatment conditions was observed and recorded with Hogland nutrient solution culture as control. The main stem height, stem-leaf fresh weight, stem-leaf dry weight, root length, root fresh weight, root dry weight, root surface area and root volume were analyzed. The relative value of plant traits of different varieties, and their correlation with salt tolerance index(STI) were determined. The results showed that the suitable NaCl concentration for salt tolerance identification by hydroponics at peanut seedling stage was 200 mmol/L, and the suitable time for plant traits determination was on the 7 th day under NaCl stress. After treated with 200 mmol/L NaCl for 7 days, the salt tolerance coefficient(STC) of Fenghua 2, Baisha 1016, Shanhua 18 and ICGV86699 was 0.77, 0.72, 0.64 and 0.53, and the STI was 0.85, 0.89, 0.60 and 0.39,respectively. The salt tolerance of the four varieties was significantly different. The correlation analysis showed that the correlation coefficient of STC,relative stem-leaf dry wight and relative root length with STI were 0.95,0.95 and 0.90,respectively, which showed a very significantly positive correlation. Therefore, the STC, relative stem-leaf dry weight, relative root length could be used as evaluation indexes of salt tolerance identification at peanut seedling stage.
