摘要
目的通过白细胞介素-4(interleukin 4,IL-4)对皮肤屏障功能和免疫反应的调控探讨马齿苋提取液治疗急性湿疹的可能机制。方法SD大鼠30只,分层随机分为3组,正常组、模型组和马齿苋组,各组10只。除正常组外其余各组采用二硝基氯苯(dinitrochlorobenzene,DNCB)建立急性湿疹模型。造模成功后,模型组和马齿苋组分别外敷1ml无菌蒸馏水和马齿苋提取液,正常组不做任何处理。用药后,采用皮肤表皮水分丧失测定仪测定各实验组大鼠经皮水分丢失量(transepidermal water loss,TEWL);ELISA技术检测大鼠血清中总免疫球蛋白E(immunoglobulin E,IgE)的含量;免疫组化法(SP法)染色IL-4和蛋白酶激活受体2(protease activated receptor-2,PAR-2)并采用病理图像分析系统半定量检测其含量。结果与正常组比,模型组大鼠IgE、IL-4、PAR-2和TEWL显著增加(P<0.01);与模型组比,马齿苋组大鼠IgE、IL-4、PAR-2和TEWL显著降低(P<0.05)。结论马齿苋提取液可通过抑制IL-4的分泌,降低IgE的释放,调节免疫反应,同时抑制PAR-2的表达,降低TEWL,恢复皮肤屏障功能,达到治疗急性湿疹的作用。
Objective Through the regulation of skin barrier function and immune response,to explore the possible mechanism of purslane in the treatment of rats with atopic eczema.Methods Forty Sprague-Dawley rats were randomly divided into normal group,model group,Huangbai group,purslane group,10 rats in each group.In addition to the normal group,the other groups used dinitrochlorobenzene(DNCB)to establish atopic eczema model.After the success of the model manufacture,1ml distilled water,1ml cortex phellodendri extracts and 1ml Portulaca extracts were applied to the model group,the Huangbai group and the purslane group respectively,twice a day for nine days.The transdermal water loss(TEWL)of rats in each experimental group was measured by skin cutaneous moisture loss tester.The content of total immunoglobulin E in serum was detected by ELISA.immunohistochemistry(SP method)to stain interleukin 4(IL-4)and protease activated receptor-2(PAR-2).The contents of IL-4 and PAR-2 were detected semiquantitatively by using pathological image analysis system.Results Compared with the normal group,the levels of IgE,IL-4,PAR-2 and TEWL in the model group were significantly increased(P<0.01).Compared with the model group,the levels of IgE,IL-4,PAR-2 and TEWL in the purslane group were significantly decreased(P<0.05).Conclusion Purslane can inhibit the secretion of IL-4,reduce the release of IgE,regulate the immune response,reduce the TEWL,inhibit the expression of PAR-2 and restore the barrier function of the skin to achieve the treatment of atopic eczema.
