刺参低聚肽的质量评价和抗疲劳作用研究 被引量:8

摘要 对刺参低聚肽的氨基酸组成、分子量分布、营养成分、微量元素、农兽药残留及急性经口毒性试验进行综合评价,研究并比较刺参低聚肽、大豆低聚肽和海参粉对小鼠的抗疲劳作用。分别灌胃不同剂量(0.085、0.17、0.50 g/kg)的刺参低聚肽、大豆低聚肽、海参粉,30 d后测定小鼠的负重游泳时间、血清尿素氮和肝糖原含量。结果表明,刺参低聚肽中氨基酸种类齐全,富含多种人体必需氨基酸,分子量低于1 000 Da低聚肽含量大于98 %,主要分布在180 Da^500 Da,含有众多的微量元素,且牛磺酸含量较高,无兽药及农药残留,无重金属超标,样品经口服的半数致死量(median lethal dose, LD50)大于10 000 mg/kg,无毒性,口服安全。与阴性对照组相比,刺参低聚肽、大豆低聚肽和海参粉都具有抗疲劳的作用,其抗疲劳能力强弱顺序为:刺参低聚肽>大豆低聚肽>海参粉,刺参低聚肽各剂量组均使得小鼠负重游泳时间明显延长,血清尿素氮含量明显降低,肝糖原含量明显增加,但并非剂量越高效果越好,最佳给药剂量为0.17 g/kg。 The amino acid composition, molecular weight distribution, nutrient composition, trace elements, drug residues and acute oral toxicity test of Apostichopus japonicus oligopeptides were evaluated comprehensively.The anti-fatigue effects of Apostichopus japonicus oligopeptides, soybean oligopeptides and sea cucumber powder on mice were studied and compared.The mice were administered by oral gavage at three different doses (0.085,0.17, 0.50 g/kg).After being fed for 30 days, the loaded swimming time, the content of serum urea nitrogen and hepatic glycogen were measured.The results showed that Apostichopus japonicus oligopeptides had different kinds of amino acids and was rich in essential amino acids.Oligopeptide with molecular weight less than 1 000 Da was more than 98 %, mainly distributed at 180 Da-500 Da.It contained many kinds of trace elements.The content of taurine was high.It didn't contain veterinary drug, pesticide residues and heavy metals.The median lethal dose (LD50) was greater than 10 000 mg/kg by oral gavage.It was no toxicity and safety.Compared with the negative control group, Apostichopus japonicus oligopeptides, soybean oligopeptides and sea cucumber powder all had anti-fatigue effects.The order was Apostichopus japonicus oligopeptides > soybean oligopeptides > sea cucumber powder.Each dose of Apostichopus japonicus oligopeptides significantly prolonged the loaded swimming time, decreased the content of serum urea nitrogen, and increased the content of hepatic glycogen significantly.However the dosage wasn't the higher the better, the best dosage was 0.17 g/kg.
